应用体外DNA转染技术已建立具有在裸鼠体内形成恶性肿瘤能力的人鼻咽癌转化细胞株。证明了人鼻咽癌DNA中含有致癌的转化基因(癌基因)。将转化细胞及操鼠肿瘤DNA经BamHⅠ、EcoRⅠ.HindⅢ、EcoRⅠ/HindⅢ等酶切后,与~(32)p-标记的Ha-ras、Ki-ras、N-ras、C-myc基因探针杂交,结果发玥与Ha-ras杂交呈现6.6kb的阳性区带,其杂交片段的长度呈现多态性,与C-myc杂交呈现8.5kb的阳性区带,而ki-ras及N-ras杂交为阴性,说明人鼻咽癌癌基因与Ha-ras、C-myc具有同源顺序。提示在细胞癌变过程中,可有几种癌基因被激活协同作用而导致癌变。本文并首次应用PCB技术,对鼻咽癌癌基因点突变激活机制进行了初步研究。
In vitro DNA transfection technology has been established with the ability to form malignant tumors in nude mice human nasopharyngeal carcinoma cell lines. The transformed cells and mouse tumor DNA were digested with BamHⅠ, EcoRⅠ.HindⅢ, EcoRI / HindⅢ, etc., and then probed with ~ (32) p-labeled Ha-ras, Ki-ras, N-ras and C-myc gene probes The results showed that the positive hybridized with Ha-ras showed a positive 6.6kb region, and the length of the hybridized fragment was polymorphic. The positive hybridized with C-myc showed a positive band of 8.5kb, while the ki-ras and N-ras Is negative, indicating that human nasopharyngeal carcinoma oncogene and Ha-ras, C-myc has a homologous sequence. Tip in the process of cancerous cells, there are several oncogenes can be activated and lead to cancer. In this paper, the first application of PCB technology, nasopharyngeal oncogene point mutation activation mechanism was studied.
