目的探讨唾液酸酶与子宫内膜癌细胞Ishikawa浸润、增殖和凋亡的关系。方法培养子宫内膜癌Ishikawa细胞,并分成对照组和处理组,处理组给予唾液酸酶活性抑制剂(DANA)处理,比对两组细胞上清液的唾液酸酶活性后通过免疫荧光检测、流式细胞学或MTT实验检测对照组和处理组细胞基质金属蛋白酶(MMPs)表达水平、细胞增殖核抗原(PCNA)水平、凋亡和增殖情况。结果处理组上清液唾液酸酶活性减弱且细胞MMPs的表达降低,处理组的细胞凋亡率明显降低,各组细胞的增殖能力无差异。结论唾液酸酶可影响Ishikawa细胞的侵袭性和凋亡率,但对细胞的增殖能力无明显影响。
Objective To investigate the relationship between sialidase and Ishikawa infiltration, proliferation and apoptosis in endometrial carcinoma cells. Methods Ishikawa cells of endometrial carcinoma were cultured and divided into control group and treatment group. The treatment group was given sialidase inhibitor (DANA) treatment. The sialidase activity of the supernatant of the two groups was compared by immunofluorescence, Flow cytometry or MTT assay was used to detect the expression of matrix metalloproteinases (MMPs), the level of proliferating cell nuclear antigen (PCNA), apoptosis and proliferation in the control and treated groups. Results In the treatment group, the sialidase activity of the supernatant was decreased and the expression of MMPs in the cells was decreased. The apoptosis rate of the treated group was significantly decreased, while the proliferative capacity of the cells in each group was not different. Conclusion Sialidase can affect the invasiveness and apoptosis rate of Ishikawa cells, but have no significant effect on the proliferation of cells.
