目的检测氧化苦参碱(Oxymatrine,OMT)对刀豆蛋白A(ConA)刺激的小鼠淋巴结T细胞增殖的影响,探讨OMT对免疫系统的作用机制,为临床用OMT治疗免疫相关性疾病提供理论和实验依据。方法利用CFDA-SE染色,流式细胞术检测淋巴细胞在多克隆刺激剂ConA和OMT的共同作同下荧光强度的变化,并应用CELLQuest软件分析OMT对小鼠淋巴结T细胞增殖的影响程度。结果500、125和31μg/mLOMT对小鼠淋巴结T细胞增殖呈剂量依赖性抑制,而16、8、4、2μg/mLOMT对小鼠淋巴结T细胞增殖起促进作用,但其剂量依赖关系不明显。结论CFDA-SE染色和流式细胞术是分析淋巴细胞增殖的有力工具;OMT对小鼠淋巴结T细胞的增殖呈双向作用。
Objective To investigate the effect of Oxymatrine (OMT) on the proliferation of mouse lymph node T cells stimulated by ConA and to explore the mechanism of action of OMT on the immune system and to provide a theoretical basis for the clinical treatment of immune related diseases with OMT. And experimental basis. Methods Using CFDA-SE staining, flow cytometry was used to detect the change of fluorescence intensity of lymphocytes under the same conditions of the polyclonal stimulators ConA and OMT. CELLQuest software was used to analyze the effect of OMT on lymphocyte T lymphocyte proliferation in mice. RESULTS: 500, 125, and 31 μg/mLOMT showed dose-dependent inhibition of proliferation of lymph node T cells, while 16, 8, 4 and 2 μg/mLOMT promoted lymph node T cell proliferation in mice, but the dose-dependent relationship was not significant. Conclusions CFDA-SE staining and flow cytometry are powerful tools for the analysis of lymphocyte proliferation; OMT has a two-way effect on the proliferation of mouse lymph node T cells.
