目的 检测妊娠高血压综合征 (妊高征 )患者胎盘组织和正常妊娠胎盘组织差异表达基因谱。 方法 Trizol法抽提组织总RNA并纯化mRNA ;应用cDNA阵列人类全基因表达谱分析芯片检测 6例妊高征患者胎盘组织和 5例正常妊娠者胎盘组织中差异表达的基因 ,扫描仪分析结果。 结果 所检测的 170 0 0个基因中 ,妊高征患者胎盘组织与正常妊娠者胎盘组织之间存在差异表达基因 96个 ,表达上调基因数为 78个 ,下调基因数为 18个。初步分析发现 ,有 85个已知功能基因 ,其中 79个功能分类基因 ,大致分为癌基因和细胞周期蛋白、细胞骨架和运动蛋白、细胞分裂、离子通道和运输蛋白、细胞凋亡和应激反应蛋白、细胞受体、免疫、细胞信号和传递蛋白、转录和翻译系统、代谢、发育、细胞分化相关基因 ,另外有 6个未分类基因、8个未知基因表达序列标签、3个cDNA片段。 结论 妊高征发病的分子机制可能涉及了一些基因表达的变化。
Objective To detect differentially expressed genes in placenta and placenta of patients with pregnancy induced hypertension (PIH). Methods Trizol method was used to extract the total RNA of tissue and to purify the mRNA. The cDNA microarray was used to detect the gene expression in the placenta from 6 cases of PIH and 5 cases of normal pregnancy. Results Among the 170 0 0 genes detected, there were 96 differentially expressed genes in placenta between PIH patients and normal pregnancies, with 78 up-regulated genes and 18 down-regulated genes. Preliminary analysis found that there are 85 known functional genes, of which 79 functional classification genes, broadly divided into oncogenes and cyclin, cytoskeleton and motor protein, cell division, ion channels and transport proteins, apoptosis and stress Reactive proteins, cellular receptors, immune, cell signaling and transfer proteins, transcription and translation systems, metabolism, development, and cell differentiation related genes. In addition there are 6 unclassified genes, 8 unknown gene expression sequence tags and 3 cDNA fragments. Conclusion The molecular mechanism of PIH may involve some changes in gene expression.
