目的观察十溴联苯醚(BDE-209)对成年大鼠海马胆碱能系统和γ-氨基丁酸(GABA)的影响。方法将64只健康SPF级SD雄性大鼠随机分为4组,溶剂对照(花生油)组和250、500、1 000 mg/kg BDE-209染毒组,每组16只。采用灌胃方式进行染毒,每天1次,连续染毒60 d。采用Morris水迷宫试验检测大鼠的学习和记忆能力;采用化学比色法测定海马组织乙酰胆碱转移酶(Ch AT)、乙酰胆碱酯酶(ACh E)的活力和乙酰胆碱(ACh)的含量,采用免疫组化法检测海马组织GABA的表达情况。结果与溶剂对照组比较,在同一训练时间,各剂量BDE-209染毒组大鼠的逃避潜伏期均延长,跨台次数均减少;海马组织内Ch AT、TCh E活力和ACh含量及CA1区、CA3区和DG内GABA含量均较低,差异有统计学意义(P<0.05)。且随着BDE-209染毒剂量的升高,大鼠的逃避潜伏期呈延长趋势,跨台次数呈下降趋势,海马组织内Ch AT、TCh E活力和ACh含量及CA1区、CA3区和DG内GABA表达均呈下降趋势。结论 BDE-209亚慢性染毒导致大鼠学习和记忆能力下降,可能与海马胆碱能系统和GABA表达下降有关。
Objective To observe the effects of decabromodiphenyl ether (BDE-209) on the cholinergic system and γ-aminobutyric acid (GABA) in the hippocampus of adult rats. Methods Sixty-four male Sprague-Dawley (SD) SD male rats were randomly divided into 4 groups, solvent control group (peanut oil) and 250, 500 and 1 000 mg / kg BDE-209 groups. Gavage by way of exposure, 1 day, continuous exposure to 60 d. The Morris water maze test was used to detect the learning and memory ability of rats. The contents of acetylcholinesterase (AChE) and acetylcholinesterase (ACh) in hippocampus were determined by chemical colorimetry. Method to detect the expression of GABA in hippocampus. Results Compared with the solvent control group, the escape latency of rats in BDE-209 treatment group was prolonged at the same training time and the number of crosses was reduced. The activities of Ch AT, TCh E and ACh in CA1, The content of GABA in CA3 area and DG was lower, the difference was statistically significant (P <0.05). And with the increase of the dose of BDE-209, the escape latency of rats increased and the number of cross-platform decreased. The activities of Ch AT, TCh E and ACh in hippocampus and the expression of CA1, CA3 and DG GABA expression showed a downward trend. Conclusion The sub-chronic exposure to BDE-209 leads to decreased learning and memory in rats, which may be related to the decrease of cholinergic system and GABA expression in the hippocampus.
