目的:观察参苏饮在炎症人支气管上皮细胞(16HBE)表达β-防御素2(h BD-2)过程中对NF-κB活性的影响。方法:选择SD雄性大鼠,灌胃给药参苏饮及其拆方,心脏采血制备含药血清;传代培养人支气管上皮细胞(16HBE);造模LPS(1 mg/L)刺激16HBE后不同时间点(3 h、6h、12 h、24 h)测定细胞增殖活性及培养上清液中TNF-α、IL-8含量,建立16HBE炎症模型。滤板法检测给药后不同时间点(0.5 h、1 h、3 h、5 h、8 h)细胞NFk B的激活情况。结果:与模型组比较,全方组、益气解表组、止咳化痰组NF-κB蛋白活性在1 h、3 h、5 h、8 h显著性升高(P<0.05)。结论:参苏饮能促进NF-κB蛋白的活化,且全方组的效应优于拆方组,这可能与增强h BD-2的转录启动有关。
Objective: To observe the effect of Shen Su Yin on the activity of NF-κB during the expression of β-defensin 2 (h BD-2) in human bronchial epithelial cells (16HBE). Methods: SD male rats were given gavage and Shenfu decoction orally, and blood was collected from the heart to prepare drug-containing serum. Human bronchial epithelial cells (16HBE) were subcultured and cultured with LPS (1 mg / L) (3 h, 6 h, 12 h, 24 h). The levels of TNF-α and IL-8 in the culture supernatant were measured to establish a model of 16HBE inflammation. The activation of NFκB in cells at different time points (0.5 h, 1 h, 3 h, 5 h, 8 h) was detected by plate filter. Results: Compared with the model group, the activity of NF-κB in Qi Fang Decoction, Yiqi Jiedu Decoction and Zhike Huatan Decoction group were significantly increased at 1 h, 3 h, 5 h, 8 h (P <0.05). Conclusion: ShenShen Decoction can promote the activation of NF-κB, and the effect of all prescriptions is better than the disassembled prescriptions, which may be related to the enhancement of transcriptional activation of h BD-2.
