目的:探讨苦丁茶水提物体外抗柯萨奇病毒B3(CVB3)的作用及机制的研究。方法:采用组织细胞培养法,通过观察细胞病变(CPE)以及四甲基偶氮唑蓝(MTT)法检测细胞活性、测定病毒抑制率,以确定苦丁茶水提物体外抗病毒活性,并采用预防、治疗和同时用药干预的方法初步探讨其抗病毒作用机制。结果:苦丁茶水提物有明显的体外抗CVB3活性,对HeLa细胞的半数中毒浓度(TC50)为45700.3μg/ml,半数抑制浓度(IC50)为119.18μg/ml,治疗指数(TI)为383.46。结论:苦丁茶水提物在体外具有明显的抑制CVB3作用。
Objective: To investigate the effect and mechanism of external anti-Coxsackievirus B3 (CVB3) in aqueous extract of Kuding tea. METHODS: Tissue culture method was used to detect cell viability and determine virus inhibition rate by observing cytopathic effect (CPE) and MTT assay to determine the anti-viral activity of Kudingcha water extract. The methods of prevention, treatment and simultaneous drug intervention were used to explore the mechanism of antiviral action. RESULTS: The aqueous extract of Kuding tea had obvious anti-CVB3 activity in vitro. The half poisoning concentration (TC50) of HeLa cells was 4570 0.3 μg/ml, the IC50 was 119.18 μg/ml, and the therapeutic index (TI) was 383.46. . Conclusion: The aqueous extract of Kuding tea has obvious inhibition of CVB3 in vitro.
