目的:通过糖尿病动物模型研究补中益气汤对胃黏膜损伤与胃中性黏蛋白、NO缺陷的影响。方法:将造模成功的糖尿病大鼠60只随机分为4组,并设正常组大鼠12只。模型B组与治疗B组在糖尿病大鼠基础上通过口服中药(青皮、枳壳、干姜、黄柏)诱导建立糖尿病阴阳两虚证动物模型,治疗A组、治疗B组均予灌服补中益气汤。观察各组大鼠胃黏膜损伤情况,采用紫外分光光度法测定胃黏膜NO代谢产物NO2-、NO3-、ONOO-及胃中性黏蛋白含量。结果:糖尿病大鼠均存在胃黏膜充血和出血情况,治疗A组胃黏膜损伤情况改善。与正常组比较,其余各组糖尿病大鼠中性黏蛋白含量均显著降低(P<0.01)。治疗A组与模型A组比较、治疗B组与模型B组比较,中性黏蛋白含量均明显下降(P<0.05)。模型A组NO2-、NO3-、ONOO-均较正常组显著降低(P<0.05,P<0.01)。治疗A组NO3-、NO2-、ONOO-均较模型A组显著升高(P<0.05,P<0.01)。模型B组NO2-、NO3-均较正常组显著降低(P<0.01),治疗B组NO2-、NO3-、ONOO-较模型B组均无显著改变(P>0.05)。结论:糖尿病状态可能损害NO相关的胃中性黏蛋白N-糖基化,导致胃黏蛋白N-聚糖缺陷、胃黏膜屏障结构破坏、胃黏膜损伤,补中益气汤对以上异常有一定的治疗作用。阴阳两虚使以上异常进一步加重,且应用补中益气汤无效。
OBJECTIVE: To study the effect of Buzhong Yiqi Decoction on gastric mucosal injury and neutrophil mucin, NO deficiency in diabetic animal models. Methods: Sixty diabetic diabetic rats were randomly divided into 4 groups and 12 normal rats. The model group B and the treatment group B were established by animal model of oral administration of Chinese herbs (Qingpi, Citrus aurantium, dried ginger and cork) on the basis of diabetic rats. Yiqi soup. Gastric mucosal lesions were observed in each group. The contents of NO2-, NO3-, ONOO- and neutrophilic mucin in gastric mucosa were measured by UV spectrophotometry. Results: Gastric mucosal hyperemia and hemorrhage were all found in diabetic rats, and gastric mucosal lesion in treatment group A was improved. Compared with the normal group, the content of neutrophil in the other groups were significantly decreased (P <0.01). Compared with model group A, the content of neutral mucin in treatment group A and B was significantly decreased (P <0.05). NO2-, NO3- and ONOO- in model group A were significantly lower than those in normal group (P <0.05, P <0.01). Treatment group A NO3-, NO2-, ONOO- were significantly higher than model group A (P <0.05, P <0.01). NO2- and NO3- in model group B were significantly lower than those in normal group (P <0.01). No significant changes were found in NO2-, NO3- and ONOO-treated group B compared with model group B (P> 0.05). CONCLUSION: The state of diabetes may impair N-glycosylation of NO related gastric mucin, resulting in the defects of gastric mucosal N-glycan defect, gastric mucosal barrier structure and gastric mucosal injury. Buzhong Yiqi Decoction has some of the above abnormalities Therapeutic effect. Yin and yang to make the above abnormalities further aggravate, and the application Bu Zhong Yi Qi Tang invalid.
