目的:比较复方鱼腥草合剂水提物、醇提物、挥发油以及完整成分对db/db小鼠肾损伤的作用,并探索其可能机制。方法:选取8只db/m小鼠为正常组;48只db/db小鼠分为模型组、盐酸二甲双胍组、复方鱼腥草合剂水提物组、醇提物组、挥发油组以及完整成分组,每组8只,给药8周,记录小鼠体质量以及血糖,8周后检测小鼠血糖、肾功能指标及肾脏指数,ELISA试剂盒法检测血清胰岛素水平、TGF-β1、FN、GLP-1、GIP水平,HE染色观察小鼠肾脏病理变化。结果:与模型组相比,醇提物、挥发油、完整成分组小鼠血清胰岛素水平、胰岛素敏感指数升高,BUN、Scr水平有所改善,差异无统计学意义。血清TGF-β1,FN,GIP水平降低(P<0.05),GLP-1含量升高(P<0.05)。肾脏组织病理改善明显,且以完整成分组改变最为显著。结论:复方鱼腥草合剂对糖尿病所致肾损伤有一定改善作用,其保护机制可能与下调TGF-β1、FN水平,减少ECM沉积,调节GLP-1、GIP分泌有关。
Objective: To compare the effects of compound Houttuynia mixture aqueous extract, alcohol extract, volatile oil and intact components on renal injury in db / db mice and to explore its possible mechanism. Methods: Eight db / m mice were selected as normal group. 48 db / db mice were divided into model group, metformin hydrochloride group, compound Houttuynia cordata extract water extract group, alcohol extract group, volatile oil group and complete components The rats in each group were sacrificed for 8 weeks. The body weight and blood glucose of mice were recorded. After 8 weeks, the blood glucose, renal function index and renal index of mice were measured. The levels of serum insulin, TGF-β1, FN, GLP-1, GIP levels, HE staining observed pathological changes in the kidneys of mice. Results: Compared with the model group, serum insulin, insulin sensitivity index and BUN and Scr levels in alcohol extract, volatile oil and intact mice had no significant difference. Serum TGF-β1, FN, GIP levels decreased (P <0.05), GLP-1 content increased (P <0.05). Kidney histopathology improved significantly, and the most significant changes in the complete component group. CONCLUSION: Compound Houttuynia Mixture can improve the renal injury caused by diabetes. The protective mechanism may be related to down-regulating the levels of TGF-β1 and FN, decreasing the deposition of ECM and regulating the secretion of GLP-1 and GIP.
