目的:探讨在应激和注射消炎痛诱导的胃溃疡大鼠胃粘膜白细胞介素8(IL8)免疫反应阳性物质含量的变化.方法:将浸水应激、注射消炎痛大鼠各自分为05,1,2和4h组,取胃窦标本进行免疫组化染色,并以图像分析方法分析各个时间点胃粘膜细胞中IL8免疫阳性物质含量的变化.结果:与正常对照组相比,在浸水应激、注射消炎痛05h时大鼠胃粘膜细胞内IL8免疫反应阳性物质含量明显降低,随着时间延长IL8含量水平逐渐回升,4h时明显高于05h时,但仍较正常对照组为低.结论:浸水应激或消炎痛性胃溃疡形成早期,胃窦部粘膜细胞内IL8一过性被释放到细胞外,同时可能由于胃粘膜细胞中IL8的合成增强,导致粘膜细胞内IL8含量水平回升.
Objective: To investigate the changes of gastric mucosal interleukin-8 (IL-8) immunoreactive substance in rats with gastric ulcer induced by indomethacin and indomethacin. Methods: Rats in water immersion stress and indomethacin injection were divided into 0, 5, 1, 2 and 4 h groups. The specimens of gastric antrum were harvested and immunohistochemically stained. The levels of IL 8 immunopositive substance content changes. Results: Compared with the normal control group, the content of IL8 immunoreactive substance in gastric mucosal cells was significantly decreased at 05h after indomethacin infusion in water immersion stress. With the prolongation of IL8 content levels, 4h was significantly higher than 0 5h, but still lower than the normal control group. Conclusion: In the early stage of water stress or indomethacin-induced gastric ulcer, IL 8 transiently is released to the extracellular of mucosal cells in gastric antrum, and may be due to the enhanced synthesis of IL-8 in gastric mucosal cells, IL 8 content levels rise.
