目的:观察消癌解毒方对Walker256大鼠T淋巴细胞亚群和自然杀伤细胞(natural kill,NK)的影响。方法:建立Walker256大鼠模型,50只雄性SD大鼠分为正常组,模型组,中药低剂量组,中药中剂量组,中药高剂量组,每组10只。造模后中药各剂量组进行中药干预,正常组,模型组未进行药物干预。在造模后第10天用流式细胞仪分别检测各组大鼠胸腺,骨髓,外周血T淋巴细胞亚群及NK细胞活性。结果:消癌解毒方干预后,可见低剂量、中剂量组大鼠胸腺指数明显上升,中剂量组大鼠脾指数明显上升,与模型组比较,差异均有显著性(P<0.05);中剂量、高剂量组大鼠骨髓、胸腺细胞与模型组相比,G0/G1期比例显著降低(P<0.05),S期比例明显升高(P<0.05);中剂量组CD4+/CD8+比值明显上升,与模型组相比,差异有显著性(P<0.05);各剂量组NK活性明显增高(P<0.01)。结论:消癌解毒方对Walker256大鼠胸腺指数、脾指数及骨髓、胸腺细胞周期有一定的调节作用,对淋巴细胞亚群CD4+、CD8+、CD4+/CD8+比值及NK细胞活性有一定的调节作用。消癌解毒方具有免疫保护作用可能是其抗癌作用机制之一。
Objective: To observe the effect of Xiaoyao Jiedu Fang on T lymphocyte subsets and natural killer (NK) in Walker256 rats. Methods: Walker256 rat model was established. 50 male SD rats were divided into normal group, model group, low dose Chinese medicine group, middle dose Chinese medicine group and high dose Chinese herb group, with 10 rats in each group. After the model was made, traditional Chinese medicine intervention was carried out for all dose groups of Chinese medicine. No drug intervention was made in normal group and model group. On the 10th day after the model establishment, the T lymphocyte subsets and NK cell activities in the thymus, bone marrow and peripheral blood were detected by flow cytometry. Results: After the intervention of Xiaoyan Jiedu Fang, the thymus index of rats in low dose and middle dose group increased obviously, while the spleen index of middle dose group increased obviously (P <0.05), the difference was significant Compared with the model group, the proportion of G0 / G1 phase was significantly decreased (P <0.05) and the proportion of S phase was significantly increased (P <0.05). The ratio of CD4 + / CD8 + (P <0.05). Compared with the model group, the NK activity in each dose group was significantly increased (P <0.01). Conclusion: Xiaoyao Jiedu Fang regulates the thymus index, spleen index and the cell cycle of bone marrow and thymus in Walker256 rats, and regulates the ratio of CD4 +, CD8 +, CD4 + / CD8 + and NK cell activity in lymphocyte subsets. Xiao Fang Jie Du Fang has an immunoprotective effect may be one of its anti-cancer mechanism.
