应用NADPH-d 酶组织化学方法,观察了大鼠烫伤后脑内NOS阳性神经元数目和阳性反应面积的变化及NGF对其影响。结果显示:大鼠体表烫伤后3 天,纹状体NOS阳性神经元数目明显增加,染色呈强阳性,阳性反应面积增加。海马的NOS阳性神经元数变化不明显,仅见阳性反应面积增加,NGF可降低纹状体的NOS阳性神经元数目、阳性反应面积,NOS阳性神经元着色较淡;海马的NOS阳性反应面积减少,NGF的作用与L-NAME抑制NOS的作用相似。这些结果提示,NGF可能通过降低NOS活性,从而减轻烫伤引起的神经元损伤。
NADPH-d enzymatic histochemistry was used to observe the changes of NOS positive neurons and the area of positive reaction after scald in rats and the effect of NGF on them. The results showed that the number of NOS positive neurons in the striatum increased significantly 3 days after scalding, the staining was strongly positive and the area of positive reaction increased. The number of NOS positive neurons in the hippocampus was not significantly changed. Only the area of positive reaction increased. NGF could reduce the number of NOS positive neurons in the striatum. The area of positive reaction and the staining of NOS positive neurons were lighter. The area of hippocampus NOS positive reaction decreased, The role of NGF is similar to that of L-NAME in inhibiting NOS. These results suggest that NGF may reduce neuronal damage induced by scald by reducing NOS activity.
