目的:探究豆梨多糖提取的技术方法和最佳工艺条件。方法:样品采用恒温水浴浸提法提取,用醇沉法得到多糖沉淀,用稀硫酸溶解后,采用苯酚-硫酸法,在波长490nm处测定吸光度,以葡萄糖标准品为对照品,计算豆梨多糖的提取率;采用正交实验法,对影响多糖提取的料液比、提取时间、提取温度等3个因素进行考察,确定豆梨多糖提取的最佳工艺条件,并对结果进行验证。结果:最佳提取工艺条件为:料液比1∶15、浸提温度95℃、浸提时间2h。此条件下得到的多糖提取率最高,平均产率为3.04%。
Objective: To explore the technical methods and the optimum technological conditions for the extraction of polysaccharides from Pear. Methods: The sample was extracted by constant temperature water bath extraction, the precipitation of polysaccharides was obtained by alcohol precipitation, dissolved with dilute sulfuric acid, the phenol-sulfuric acid method was used, the absorbance was measured at the wavelength of 490nm, glucose standard was used as reference substance, The orthogonal test was used to investigate the factors influencing the ratio of solid to liquid extraction, the extraction time and the extraction temperature of polysaccharides. The optimum extraction conditions were determined and the results were verified. Results: The optimum extraction conditions were: solid-liquid ratio 1:15, extraction temperature 95 ℃, extraction time 2h. Under this condition, the polysaccharide yield was the highest with the average yield of 3.04%.
