高压氧对帕金森病模型大鼠认知能力、脑纹状体多巴胺及其代谢产物的影响

目的:探究高压氧(HBO)处理对帕金森病(PD)模型大鼠认知能力、脑纹状体多巴胺(DA)及其代谢产物的影响。方法:选用18只健康雄性SD大鼠,按照数字表法随机分为假手术组、模型组和实验组,每组6只。6-羟基多巴胺(6-OHDA)注射用于构建大鼠PD模型,HBO处理(仅实验组)后应用水迷宫实验和Y迷宫实验检测大鼠的认知能力;利用高效液相色谱测定各组大鼠大脑纹状体DA、高香草酸(HVA)和二羟苯乙酸(DOPAC)含量。结果:与假手术组比较,模型组大鼠逃避潜伏期所用时间明显增加,穿越平台次数和自发交替率降低(n P<0.05),DA、HVA、DOPAC含量及DA/DOPAC、DA/HVA值均有不同程度下降(n P<0.05);实验组大鼠经过HBO处理后上述指标均有明显改善(n P<0.05)。n 结论:HBO处理能够明显提高PD模型大鼠的认知能力,逆转DA及其代谢产物异常,为临床治疗和研究提供借鉴。“,”Objective:To explore the effects of hyperbaric oxygen (HBO) on the cognitive ability, striatal dopamine (DA) level and its metabolite of rat models with Parkinson’s disease (PD).Methods:A total of 18 PD rats were randomly divided into sham group, model group, and experimental group, with 6 rats per group. The 6-OHDA injection was used to construct the PD model in all rats, while only the rats in the experimental group were given HBO treatment. Morris water maze test and Y-maze test were used to explore the cognitive ability of rats. High-performance liquid chromatography was used to determine the levels of DA, homovanillic acid (HVA) and dihydroxy-phenylacetic acid (DOPAC) in rats.Results:Compared with the sham group, the escape latency was significantly increased, while the times of rats crossing the platform and the spontaneous alternation rate were decreased in the model group (n P<0.05); in addition, the contents of DA, HVA, DOPAC and the ratios of DA/DOPAC and DA/HVA in the model group were all decreased to different degrees compared with those in the sham group, all showing statistically significant differences (n P<0.05); and the above indexes in the experimental group were significantly improved after hyperbaric oxygen treatment, and the differences were statistically significant (n P<0.05).n Conclusion:Hyperbaric oxygen therapy can obviously improve the cognitive ability of PD rats and DA and its metabolites, thus providing reference for clinical therapy and research.
