目的:评价维拉帕米(verapamil,Ver)对人巩膜成纤维细胞(humanscleralfibroblasts,HSF)增殖的影响。方法:培养的HSF分别加入不同浓度的Ver及5-FU、肝素(heparin,Hep)、地塞米松(dexamethasone,Dex),用四甲基偶氮唑盐(MTT)比色法,在酶联免疫测定仪上测540nm处吸光值,计算出各药物浓度条件下的净增抑制率。实验数据行t检验。结果:Ver在20mg/L以上使细胞增殖率显著下降;60mg/L以上使部分细胞生存受到影响。将Ver分别与不同浓度的5-FU、Hep、Dex合用,抗增殖作用明显增强。结论:Ver对HSF增殖有抑制作用,并能增强5-FU、Hep、Dex的抗增殖作用效果
Objective: To evaluate the effect of verapamil (Ver) on the proliferation of human scleral fibroblasts (HSF). Methods: Cultured HSF were added with different concentrations of Ver and 5-FU, heparin (Hep), dexamethasone (Dex) and MTT colorimetric method respectively. Measured at 540nm absorbance measured on an immunoassay to calculate the net increase inhibition rate of each drug concentration conditions. Experimental data line t test. Results: Ver at 20mg / L or more significantly reduced cell proliferation rate; 60mg / L above the part of cell survival affected. The combination of Ver with 5-FU, Hep and Dex in different concentrations significantly enhanced the antiproliferative effect. Conclusion: Ver has an inhibitory effect on the proliferation of HSF and enhances the antiproliferative effects of 5-FU, Hep and Dex
