目的通过单个胚胎荧光定量RT-PCR的方法分析不同质量和来源的胚胎与IGF-Ⅱ表达量之间的关系。方法收集不适合移植及冷冻保存的胚胎,收集胚胎评分资料和所属患者资料。运用荧光定量RT-PCR方法测定其IGF-Ⅱ表达量。结果人类植入前胚胎形态学评分越高,其IGF-ⅡmRNA的表达量越高;年龄较轻和妊娠的病人中其胚胎中IGF-Ⅱ的基因表达量较高。结论荧光定量RT-PCR技术可用于检测单个植入前胚胎的mRNA含量;IGF-II可作为胚胎质量的标记物;IGF-II基因表达量和胚胎的形态学评分呈正相关。
Objective To analyze the relationship between the expression of IGF-Ⅱ and embryos of different quality and origin by single quantitative RT-PCR. Methods The embryos not suitable for transplantation and cryopreservation were collected and the data of embryo scoring and the patients’ information were collected. The expression of IGF-Ⅱ was measured by fluorescence quantitative RT-PCR. Results The higher the morphological score of preimplantation embryos was, the higher the expression of IGF-ⅡmRNA was. The lighter the gestational age, the higher the gene expression of IGF-Ⅱ was in embryos. Conclusion Fluorescent quantitative RT-PCR can be used to detect the mRNA content of single preimplantation embryos. IGF-II can be used as a marker of embryo quality. The expression of IGF-II gene is positively correlated with embryo morphological score.
