目的 研究三氧化二砷( As2 O3 )对结肠癌细胞的抑制作用。方法 以结肠癌 Lo Vo 细胞株作为研究对象,通过 M T T 法测定 As2 O3 对 Lo Vo 细胞的增殖抑制效应,采用吖啶橙荧光染色、 Td T 介导的 D N A 末端标记法及流式细胞仪检测药物作用前后细胞在形态学及细胞周期分布等方面的变化。结果 M T T 显示 As2 O3 能抑制 Lo Vo 细胞的增殖,这种抑制作用呈现一定的时间、剂量依赖关系。形态学观察发现 As2 O3 诱导的 Lo Vo 细胞死亡呈现凋亡特征。 As2 O3 在低浓度时主要干扰细胞在 S期的通过,高浓度时则选择性诱导 S期细胞凋亡。结论 As2 O3 可能是通过诱导细胞凋亡而抑制 Lo Vo 细胞增殖,这种杀伤作用存在周期特异性,这一结论可能有助于治疗方案的合理设计。
Objective To study the inhibitory effect of arsenic trioxide (As2O3) on colon cancer cells. Methods Colon cancer Lo Vo cell line was used as the research object. The inhibitory effect of As2O3 on the proliferation of Lo Vo cells was measured by M T T method. Acridine orange fluorescence staining, Td T-mediated D N A end labeling and flow were used. Cytometry was used to detect changes in cell morphology and cell cycle distribution before and after drug treatment. Results M T T showed that As2O3 could inhibit the proliferation of Lo Vo cells. This inhibition showed a certain time- and dose-dependent relationship. Morphological observations revealed that As2O3-induced Lo Vo cell death exhibited apoptotic features. As2O3 mainly interferes with the passage of cells at S phase at low concentrations, and selectively induces S phase cell apoptosis at high concentrations. Conclusions As2O3 may inhibit the proliferation of Lo Vo cells by inducing apoptosis, and this killing effect is cycle-specific. This conclusion may be helpful for the rational design of therapeutic protocols.
