脂多糖对人脐静脉内皮细胞凋亡的影响及意义

目的 :探讨脂多糖 (LPS)对血管内皮细胞 (VEC)损伤的机制以及在急性肺损伤 (ALI)发病中的作用。方法 :通过建立人脐静脉内皮细胞体外培养 ,采用流式细胞技术观察LPS对人脐静脉内皮细胞 (HUVEC)凋亡的影响。结果 :LPS加入细胞培养中 ,可明显诱导HUVEC凋亡 ,并随着作用时间的延长和LPS浓度增加 ,细胞凋亡率也相应增加 ,一定范围内呈时间、剂量、效应关系。结论 :细胞凋亡是血管内皮细胞受损的主要方式之一。

Objective: To investigate the mechanism of lipopolysaccharide (LPS) on vascular endothelial cells (VEC) injury and its role in the pathogenesis of acute lung injury (ALI). Methods: Human umbilical vein endothelial cells were cultured in vitro and the effects of LPS on the apoptosis of human umbilical vein endothelial cells (HUVECs) were observed by flow cytometry. Results: The apoptosis of HUVEC induced by LPS was obviously induced by LPS, and with the prolongation of action time and the increase of LPS concentration, the rate of apoptosis increased correspondingly, and the time, dose and effect were all within certain range. Conclusion: Apoptosis is one of the main ways of vascular endothelial cell damage.
