目的观察慢性氟中毒大鼠骨组织中激动型G蛋白(Gs)和抑制型G蛋白(Gi)含量的变化。方法将48只断乳2周的Wistar大鼠按体重随机分为4组,分别为对照(自来水)组和50、150、250 mg/L NaF染毒组,每组12只,雌雄各半。采用自由饮用的方式进行染毒,连续染毒6个月。检测大鼠尿氟、骨氟含量及氟斑牙发生率以及骨组织中Gs和Gi的水平。结果与对照组比较,各剂量NaF染毒组大鼠骨氟含量及雄性大鼠尿氟含量以及150、250 mg/L NaF染毒组雌性大鼠尿氟含量均较高,各剂量NaF染毒组大鼠氟斑牙的检出率也均较高,仅150 mg/L NaF染毒组雌性大鼠骨组织中Gs含量以及雄性大鼠骨组织中Gs、Gi含量均较高,差异有统计学意义(P<0.05);各剂量NaF染毒组大鼠骨组织中Gi/Gs值无明显变化。随着NaF染毒剂量的升高,大鼠的尿氟、骨氟含量均呈上升趋势;而骨组织中Gs、Gi含量均呈先升高后下降的趋势。对照组大鼠无氟斑牙发生,而各剂量NaF染毒组大鼠氟斑牙的检出率均为100%。结论 G蛋白在氟骨症的形成过程中可能发挥一定的促进作用。
Objective To observe the changes of the contents of Gs and Gi in chronic fluorosis rats. Methods Wistar rats of 48 weaned 2 weeks were randomly divided into 4 groups according to body weight. With the increase of NaF dose, urinary fluoride and bone fluoride content of rats showed an upward trend. However, the content of Gs and Gi in bone increased first and then decreased. In the control group, the incidence of dental fluorosis was 100% in rats exposed to NaF. Conclusion G protein may play a promoting role in the formation of skeletal fluorosis.
