目的:探讨FLIP、FADD和Caspase-8在人骨肉瘤组织及骨软骨瘤组织中的表达及其与骨肉瘤细胞凋亡的关系。方法:应用免疫组化法检测FLIP、FADD和Caspase-8蛋白在43例骨肉瘤组织及30例骨软骨瘤组织中的表达,并经图像分析系统检测FLIP、FADD和Caspase-8蛋白的平均光密度(OD)值。用激光扫描共聚焦显微镜检测骨肉瘤及骨软骨瘤中细胞凋亡分布情况并对各因子之间进行相关性分析。结果:FLIP蛋白阳性物质在骨肉瘤中的表达高于骨软骨瘤,P<0.05;而FADD和Caspase-8蛋白在骨肉瘤中的表达低于骨软骨瘤,P<0.05。FLIP和Caspase-8蛋白在骨肉瘤中表达呈负相关,r=-0.843,P<0.05;而FLIP、FADD蛋白在骨肉瘤中表达无相关性。骨肉瘤组细胞凋亡指数(AI)显著低于骨软骨瘤组,P<0.05;FLIP蛋白表达与细胞凋亡指数呈负相关,r=-0.726,P<0.05;而Caspase-8蛋白表达与骨肉瘤细胞凋亡呈正相关,r=0.446,P<0.05;FADD蛋白表达与与凋亡指数无相关性。结论:FLIP蛋白对骨肉瘤细胞凋亡有抑制作用,Caspase-8蛋白在骨肉瘤中呈低表达,抑制了肿瘤细胞凋亡。如能抑制FLIP的表达,促进Caspase-8的表达,可提高骨肉瘤细胞对凋亡的敏感性。
Objective: To investigate the expression of FLIP, FADD and Caspase-8 in human osteosarcoma and osteochondroma and their relationship with apoptosis of osteosarcoma. Methods: The expression of FLIP, FADD and Caspase-8 protein in 43 osteosarcoma tissues and 30 osteochondroma tissues were detected by immunohistochemistry. The average fluorescence intensity of FLIP, FADD and Caspase-8 protein was detected by image analysis system Density (OD) value. Laser scanning confocal microscopy was used to detect the distribution of apoptosis in osteosarcoma and osteochondroma and to analyze the correlation between the factors. Results: The expression of FLIP protein in osteosarcoma was higher than that in osteochondroma (P <0.05). The expression of FADD and Caspase-8 protein in osteosarcoma was lower than that in osteochondroma (P <0.05). There was a negative correlation between FLIP and Caspase-8 protein expression in osteosarcoma, r = -0.843, P <0.05; however, there was no correlation between FLIP and FADD protein expression in osteosarcoma. Osteosarcoma group apoptosis index (AI) was significantly lower than osteochondroma group, P <0.05; FLIP protein expression and apoptosis index was negatively correlated, r = -0.726, P <0.05; and Caspase-8 protein expression and Osteosarcoma cell apoptosis was positively correlated, r = 0.446, P <0.05; FADD protein expression and apoptosis index no correlation. Conclusion: The FLIP protein can inhibit the apoptosis of osteosarcoma cells. The expression of Caspase-8 protein is low in osteosarcoma, which inhibits the apoptosis of tumor cells. If it can inhibit the expression of FLIP and promote the expression of Caspase-8, it can increase the sensitivity of osteosarcoma cells to apoptosis.
