目的:检测大鼠在癫痫发生过程中不同时间点海马肌细胞增强因子(myocyte enhancer factor,MEF2)mRNA及细胞周期素依赖性激酶5(cyclin-dependent kinase 5,CDK5)mRNA的表达水平,并分析两者之间的相关性。方法:采用锂-匹鲁卡品腹腔注射,建立大鼠慢性癫痫模型,提取海马组织中的总RNA,并逆转录为cDNA,应用Real-time PCR检测MEF2及CDK5 mRNA的表达量。结果:MEF2A/2D mRNA于造模中止后1 d急剧下降,3 d开始上升,至4 w达最高,后渐恢复至正常水平;CDK5 mRNA的表达量在整个癫痫发生过程中均呈上升趋势,4 w达最大,6 w开始下降,8 w恢复至正常水平。结论:MEF2与CDK5均参与了慢性癫痫的形成过程,但两者的表达量在时间上无明显关联性。
AIM: To detect the expression of myocyte enhancer factor (MEF2) mRNA and cyclin-dependent kinase 5 (CDK5) mRNA in hippocampus of rats at different time points during epilepsy The correlation between the two. Methods: Chronic epilepsy model was induced by intraperitoneal injection of lithium-pilocarpine in rats. Total RNA was extracted from the hippocampus and reverse transcribed into cDNA. Real-time PCR was used to detect the expression of MEF2 and CDK5 mRNA. Results: MEF2A / 2D mRNA decreased sharply on the 1st day after discontinuation of the model and began to rise on the 3rd day, reaching the highest level on the 4th day and then returned to normal level. The expression of CDK5 mRNA showed an upward trend throughout the epileptogenesis, 4 w reached the maximum, 6 w began to decline, 8 w returned to normal levels. Conclusion: Both MEF2 and CDK5 are involved in the process of chronic epilepsy. However, the expression of MEF2 and CDK5 have no obvious correlation in time.
