目的探讨黄芩素对骨肉瘤细胞增殖、侵袭的影响及其相关机制。方法以骨肉瘤MG-63细胞为研究对象,分别加入20μL的培养液(对照组)或(0、100、200)μmol/L黄芩素溶液处理细胞48h,应用MTT法分析细胞增殖情况,通过TranswellTM侵袭实验检测细胞侵袭能力,荧光实时定量PCR测定ezrin mRNA表达,采用Western blot法检测ezrin蛋白及磷酸化ezrin(p-ezrin)蛋白表达,利用原位末端标记法(TUNEL)测定肿瘤细胞凋亡指数(AI)。结果 (100、200)μmol/L黄芩素组细胞增殖抑制率较0μmol/L黄芩素组升高,且200μmol/L黄芩素组细胞增殖抑制率明显高于100μmol/L黄芩素组。随着黄芩素浓度的增加,平均穿膜细胞数以及ezrin mRNA、ezrin蛋白、p-ezrin蛋白表达水平逐渐降低,而AI逐渐增加,与对照组和0μmol/L黄芩素组比较,差异均有显著降低,而0μmol/L黄芩素组与对照组相比无明显变化。结论黄芩素能够抑制骨肉瘤MG-63细胞增殖、侵袭,其机制可能与抑制ezrin蛋白表达和活性及促进肿瘤细胞凋亡有关。
Objective To investigate the effects of baicalein on proliferation and invasion of osteosarcoma cells and its related mechanisms. Methods The osteosarcoma MG-63 cells were treated with 20μL of culture medium (control group) or (0,100,200) μmol / L baicalein solution for 48h respectively. MTT assay was used to analyze the cell proliferation. TranswellTM The invasion ability of cells was detected by invasion assay. The expression of ezrin mRNA was detected by real-time fluorescence quantitative PCR. The expression of ezrin protein and ezrin protein were detected by Western blot. TUNEL assay was used to detect the apoptosis index (AI). Results The inhibitory rate of proliferation of (100,200) μmol / L baicalein group was higher than that of 0 μmol / L baicalein group, and the inhibitory rate of proliferation in 100 μmol / L baicalein group was significantly higher than that of 100 μmol / L baicalein group. With the increase of baicalein concentration, the average number of transmembrane cells and the expression of ezrin mRNA, ezrin protein and p-ezrin protein gradually decreased, while AI gradually increased, compared with the control group and 0μmol / L baicalein group, the differences were significant Decreased, while 0μmol / L baicalein group compared with the control group no significant change. Conclusion Baicalein can inhibit the proliferation and invasion of osteosarcoma MG-63 cells, which may be related to the inhibition of ezrin protein expression and activity and the promotion of tumor cell apoptosis.
