目的 研究药物作用前后人高转移肺癌细胞中转移相关基因表达的改变,进一步揭示淫羊藿甙(ICA)及济南假单胞菌制剂PJA 抗转移作用的机制。方法 流式细胞术及逆转录PCR(RT-PCR)技术检测转移相关基因c-m yc、Tiam -1、nm 23-H1 蛋白及m RNA 水平的变化。结果 经PJA 及ICA 处理48 h 后,PG 细胞 c-m yc、Tiam -1 基因m RNA均有不同程度地降低,而nm 23-H1 m RNA水平有不同程度地升高,药物处理96 h 后,基因水平的变化已反映至其表达产物-蛋白水平上。c-m yc及nm 23-H1 基因蛋白的升降趋势与其m RNA 水平相一致(P< 0.005),且两种水平的变化均以协同作用组最显著。结论 PJA、ICA 通过抑制转移诱导基因c-m yc、Tiam -1的表达,促进转移抑制基因nm 23的表达而发挥抗转移的作用
Objective To study the changes of metastasis-associated gene expression in human high-metastatic lung cancer cells before and after drug treatment, and further reveal the mechanism of anti-metastasis effect of Epimedium (ICA) and P. aeruginosa PJA. Methods Flow cytometry and reverse transcription PCR (RT-PCR) were used to detect the changes of c-m yc, Tiam-1, nm 23-H1 and m RNA levels of metastasis-associated genes. Results After treatment with PJA and ICA for 48 h, m RNA of cm yc and Tiam-1 gene in PG cells decreased to varying degrees, while the level of nm23-H1 m RNA increased to different extent. After 96 h of drug treatment, the gene Changes in levels have been reflected in their expression product-protein levels. The up-down tendency of c-m yc and nm23-H1 protein was consistent with its m RNA level (P<0.005), and the changes of both levels were most significant in the synergistic group. Conclusion PJA and ICA may play an anti-metastatic effect by inhibiting the expression of metastasis-inducible genes c-m yc and Tiam-1 and promoting the expression of metastasis suppressor gene nm23.
