目的:建立从大叶钩藤(Uncaria macrophylla Wall.)中制备柯诺辛碱对照品的方法。方法 :采用萃取、硅胶柱层析和重结晶方法对氨水碱后的大叶钩藤二氯甲烷提取物进行分离纯化,通过MS、IR、1H-NMR和13C-NMR进行结构鉴定,通过TLC法和HPLC法对对照品进行纯度检测。结果 :从大叶钩藤中分离纯化出柯诺辛碱对照品,质量分数>98.0%。结论:该方法制备的柯诺辛碱对照品符合中药化学对照品的相关要求,可作为钩藤药材及其制剂质量控制用的化学对照品。
Objective: To establish a method of preparing kenoconsin reference substance from Uncaria macrophylla Wall. Methods: The aqueous extract of Rhynchophyllum macrophylla was separated and purified by extraction, silica gel column chromatography and recrystallization. The structures were identified by MS, IR, 1H-NMR and 13C-NMR. And HPLC method for the purity of the reference substance. Results: The kenotine base reference substance was isolated and purified from Uncaria macrophylla with mass fraction> 98.0%. Conclusion: The konoki alkali reference substance prepared by this method conforms to the relevant requirements of the Chinese traditional medicine reference substance, and can be used as a chemical reference substance for the quality control of Uncaria rhynchophylla and its preparation.
