目的探讨参芪扶正注射液对小鼠肝转移癌组织基因表达谱的影响。方法20只小鼠采用将结肠癌C26细胞株瘤细胞液0.2mL注射于脾脏建立肝转移癌模型,随机分为两组,造模后5日,分别给予参芪扶正注射液(参芪组)和生理盐水(模型组)腹腔内注射,隔日1次,共5次;造模后15日处死动物留取检测标本,应用小鼠全基因芯片分析两组肝癌组织基因表达谱。结果(1)造模成功率100%;(2)基因表达:与模型组比较,参芪组上调基因123个,其中Ensemble数据库登录52个;下调基因1个,其中Ensemble数据库登录0个。结论参芪扶正注射液主要是通过上调多种基因促进肿瘤细胞凋亡和稳定染色体而发挥抗肿瘤作用。
Objective To investigate the effect of Shenqi Fuzheng Injection on the gene expression profile of liver metastasis in mice. Methods Twenty mice were injected with 0.2 mL tumor cell solution of colon cancer cell line C26 into the spleen to establish a model of liver metastasis and were randomly divided into two groups. After 5 days of modeling, Shenqi Fuzheng injection (Shenqi group) And normal saline (model group) were intraperitoneally injected once every other day for 5 times. Animals were sacrificed on the 15th day after the model was established, and the gene expression profiles of the two groups were analyzed by whole-genome microarray. Results (1) The success rate of model making was 100%. (2) Gene expression: Compared with model group, 123 genes were up-regulated in Shenqi group, including 52 in Ensemble database and 1 in down-regulated gene, including 0 in Ensemble database. Conclusion Shenqi Fuzheng injection can exert anti-tumor effects mainly by up-regulating multiple genes and promoting apoptosis of tumor cells and stabilizing chromosomes.
