目的探讨大鼠创伤弧菌(VV)脓毒症时肝细胞Bcl-2 mRNA、Bax mRNA表达量的动态变化情况。方法制作大鼠VV脓毒症模型(VV组),RT-PCR法测定染菌后各时间点大鼠肝细胞Bcl-2 mRNA、Bax mRNA表达量。结果感染创伤弧菌后大鼠肝细胞Bcl-2 mRNA表达量明显下降(P﹤0.05);Bax mRNA其表达量明显增加(P﹤0.05),染菌后2 h明显增高,在染菌16 h的表达量仍明显高于正常水平(P﹤0.05)。结论创伤弧菌脓毒症可致肝组织中Bcl-2 mRNA表达量下降,Bax mRNA表达量增加,早期明显增高,并持续维持在高水平。
Objective To investigate the dynamic changes of Bcl-2 mRNA and Bax mRNA expression in hepatocytes induced by Vibrio vulnificus (VV) in rats. Methods VV sepsis model was established in rats (VV group). The expression of Bcl-2 mRNA and Bax mRNA in rat hepatocytes were detected by RT-PCR at different time points. Results The expression of Bcl-2 mRNA in Vibrio vulnificus was significantly decreased (P <0.05), the expression of Bax mRNA was significantly increased (P <0.05), and significantly increased at 2 h after infection with Vibrio vulnificus. The expression level was still significantly higher than the normal level (P <0.05). Conclusions Vibrio vulnificus can induce the decrease of Bcl-2 mRNA expression and the increase of Bax mRNA expression in liver tissues. The expression of Bcl-2 mRNA in liver tissues was significantly increased at early stage and maintained at a high level.
