目的研究从玉郎伞中首次分离的2种查尔酮单体2,6-二甲氧基查尔酮(YLSA)和17-甲氧基-7-羟基-苯骈呋喃查尔酮(YLSB)对心肌细胞凋亡的作用。方法原代培养大鼠心肌细胞,利用终浓度为100μmol·L-1的过氧化氢孵育16h建立凋亡模型;ELISA法测定心肌细胞培养上清液中TNF-α含量,Annex V-FITC/PI双染法检测心肌细胞凋亡,免疫组化法检测NF-κBp65、Bcl-2、Bax蛋白的表达。结果YLSA和YLSB均可抑制心肌细胞凋亡,降低TNF-α活性,下调NF-κBp65和Bax蛋白表达,上调Bcl-2蛋白表达,增加Bcl-2/Bax比值。结论YLSA和YLSB具有抑制心肌细胞凋亡的作用。
Objective To investigate the effects of two kinds of chalcone monomer, 2,6-dimethoxy-chalcone (YLSA) and 17-methoxy-7-hydroxy-benzofuran chalcone (YLSB ) On cardiomyocyte apoptosis. Methods Primary cultured rat cardiomyocytes were cultured in the presence of hydrogen peroxide at a final concentration of 100 μmol·L-1 for 16 h to establish a model of apoptosis. ELISA was used to determine the content of TNF-α in the supernatant of cardiomyocytes. Annex V-FITC / PI Cardiomyocytes apoptosis was detected by double staining. The expressions of NF-κBp65, Bcl-2 and Bax protein were detected by immunohistochemistry. Results Both YLSA and YLSB could inhibit the apoptosis of cardiomyocytes, decrease the activity of TNF-α, down-regulate the expressions of NF-κBp65 and Bax, up-regulate the expression of Bcl-2 and increase the ratio of Bcl-2 / Bax. Conclusion YLSA and YLSB can inhibit cardiomyocyte apoptosis.
