目的:建立以反相高效液相色谱法测定人血浆中莫西沙星浓度的方法。方法:色谱柱为Gemini C18,固定进样环为20μL,流动相为甲醇-水-三乙胺(112.5∶208.9∶0.06),流速为1mL·min-1,紫外检测波长为289nm,柱温为25℃,灵敏度为0.01AUFS。结果:莫西沙星检测浓度在0.3125~10μg·mL-1范围内线性关系良好(r=0.9998);其低、中、高3种浓度(10、2.5、0.625μg.mL-1)的回收率分别为99.46%、103.12%、96.14%;日内RSD分别为3.27%、3.35%、0.92%,日间RSD分别为7.19%、6.25%、6.68%。结论:本方法简单、快速、准确、灵敏度高,可用于莫西沙星的血药浓度测定及药动学研究。
Objective: To establish a method for the determination of moxifloxacin in human plasma by reversed-phase high performance liquid chromatography. Methods: The chromatographic column was Gemini C18 with the fixed injection loop of 20μL, mobile phase of methanol-water-triethylamine (112.5:208.9:0.06), flow rate of 1mL · min-1, UV detection wavelength of 289nm, column temperature 25 ℃, the sensitivity of 0.01AUFS. Results: The calibration curve of moxifloxacin was linear in the range of 0.3125-10μg · mL-1 (r = 0.9998). The recoveries of moxifloxacin at low, medium and high concentrations (10,2.5,0.625μg.mL-1) RSD were 3.27%, 3.35% and 0.92% respectively, and the intraday RSD were 7.19%, 6.25% and 6.68% respectively. Conclusion: The method is simple, rapid, accurate and sensitive. It can be used for the determination of moxifloxacin and pharmacokinetics.
