【目的】利用SSR标记进行毛葡萄遗传多样性的分析,阐明毛葡萄种质的亲缘关系,为有效利用野生毛葡萄种质及挖掘优良基因提供参考。【方法】利用筛选的多态性SSR引物对70份毛葡萄进行SSR扩增,评价不同种质遗传多样性,并分析亲缘关系。【结果】从40对SSR引物中筛选出16对多态性引物,共扩增出123条带,每对引物可检测到等位位点数3~15个,多态率26.7%~100.0%。PopGene分析结果表明,毛葡萄种群Nei’s基因多样性平均指数为0.4412,多样性信息平均指数为0.6308,具有较高的遗传多样性。Nei’s相似性系数分析结果表明,70份毛葡萄种质的遗传相似系数在0.60~0.89。UPGMA聚类分析结果表明,在遗传相似系数0.61处,70份毛葡萄材料可分为3大类群:第一类包含两份毛葡萄材料,编号为70和91,均来自广西罗城县;第二类包含5份材料,编号为28、1、129、3和131,除131外,其余4份来自广西罗城县;其他63份归为第三类,其中50份为广西罗城县毛葡萄。【结论】广西罗城县毛葡萄具有丰富的遗传多样性,可为毛葡萄种质资源的利用和品种选育提供参考。
【Objective】 The genetic diversity of Gossypium hirsutum L. was analyzed by using SSR markers to elucidate the genetic relationship of Gossypium hirsutum germplasm resources and provide references for the effective utilization of Gossypium hirsutum germplasm and the exploration of excellent genes. 【Method】 SSR primers were used to amplify SSR primers of 70 accessions to evaluate the genetic diversity of different germplasms and to analyze the genetic relationship. 【Result】 Sixteen polymorphic primers were screened from 40 pairs of SSR primers. A total of 123 bands were amplified. The number of alleles per primer pair was 3-15 and the polymorphic rate was 26.7% -100.0%. PopGene analysis showed that the average index of Nei’s gene diversity of wool grape population was 0.4412 and the average index of diversity information was 0.6308 with high genetic diversity. Nei’s similarity coefficient analysis showed that the genetic similarity coefficients of 70 accessions were between 0.60 and 0.89. UPGMA cluster analysis showed that 70 genetic material of G. gramineus could be divided into three groups at genetic similarity coefficient of 0.61: the first type contained two copies of wool grape material numbered 70 and 91, both from Guangxi Zhuang Autonomous Region; The second category consists of five materials numbered 28, 1, 129, 3 and 131, with the exception of 131, with the remaining four being from Lucheng County, Guangxi; the other 63 are classified as Category III, of which 50 are from Guangxi Luocheng County grape. 【Conclusion】 The wool grapes in LuoCheng County of Guangxi Province are rich in genetic diversity and may provide references for the utilization of germplasm resources and the breeding of varieties.
