本文克隆了虫害诱导的水稻脂氧合酶基因OsRCI-1全编码序列,实时定量QRT-PCR检测结果表明二化螟Chilo suppressalia Walker取食能快速且持续地诱导OsRCI-1基因的表达。利用农杆菌介导的水稻遗传转化获得了两株OsRCI-1基因的RNAi沉默突变体,突变体株系ir-rci-1和ir-rci-2中目的基因OsRCI-1的表达被明显的抑制,其表达水平分别仅为对照(秀水11)的47.73%和32.33%。生测结果表明OsRCI-1基因沉默导致水稻对二化螟抗性显著地降低,取食RNAi突变体株系的二化螟体重分别是取食秀水11的1.58倍和2.15倍。因此,水稻OsRCI-1基因正调控对二化螟的抗性,该基因介导的防御途径可能在水稻虫害诱导防御反应中起重要的作用。
The full-length OsRCI-1 gene was cloned from insect-pests. The results of quantitative real-time QRT-PCR showed that Chilo suppressalia Walker could induce the expression of OsRCI-1 gene rapidly and continuously. RNAi silencing mutants of two OsRCI-1 genes were obtained by Agrobacterium-mediated rice genetic transformation. The expression of OsRCI-1 gene in the mutant lines ir-rci-1 and ir-rci-2 was significantly inhibited , The expression levels were only 47.73% and 32.33% of the control (Xiushui 11) respectively. The results of bioassay indicated that the resistance of rice to S. Chilo suppressalis was significantly reduced by the silencing of OsRCI-1 gene. The stem borer weights of the RNAi mutant strains were 1.58 and 2.15 times higher than that of Xiushui 11 respectively. Therefore, the rice OsRCI-1 gene is regulating the resistance to Chilo suppressalis, and the gene-mediated defense pathway may play an important role in rice defense-induced defense.
