【目的】研究冠心苏合丸对缺血再灌注(ischemia reperfusion injury,IRI)损伤心肌细胞的保护作用,探讨其作用机制。【方法】培养乳鼠心肌细胞,建立缺血再灌注损伤细胞模型,采用血清药理学方法,药物血清组给予体积分数10%的含药血清处理。观察计数心肌细胞搏动频率;测定各组培养液中乳酸脱氢酶(LDH)、肌酸激酶(CK)、超氧化物歧化酶(SOD)、丙二醛(MDA)的含量及细胞半胱天冬蛋白酶(Caspase-3)活性;采用流式细胞仪测定细胞凋亡率。【结果】冠心苏合丸可显著提高缺血再灌注损伤心肌细胞的搏动频率,降低细胞培养液中的LDH、CK及MDA含量,提高SOD的含量,降低心肌细胞Caspase-3活性及细胞凋亡率(均P<0.05或P<0.01)。【结论】冠心苏合丸可能通过抑制Caspase-3活性及细胞凋亡发挥对心肌缺血再灌注损伤的保护作用。
【Objective】 To investigate the protective effect of Guanxin Suhema on myocardial cells injured by ischemiareperfusion injury (IRI) and its mechanism. 【Methods】 Cardiomyocytes were cultured and the model of ischemia-reperfusion injury was established. Serum pharmacology method was used. Serum containing 10% The frequency of cardiomyocyte pulsation was observed and counted. The contents of lactate dehydrogenase (LDH), creatine kinase (CK), superoxide dismutase (SOD), malondialdehyde (MDA) Caspase-3 activity was detected by flow cytometry. 【Results】 Guanxin Suhe Pills can significantly increase the frequency of myocardial cell beating ischemia-reperfusion injury, reduce the content of LDH, CK and MDA in the cell culture medium, increase the content of SOD and the activity of Caspase-3 and the apoptosis of cardiomyocytes Mortality (all P <0.05 or P <0.01). 【Conclusion】 Guanxin Suhe Pills may exert a protective effect on myocardial ischemia-reperfusion injury by inhibiting Caspase-3 activity and apoptosis.
