目的:观察理肺汤对哮喘大鼠肺病理损伤及血清Th1/Th2亚群失衡的影响。方法:将48只SD雄性大鼠随机分为正常组(A组),模型组(B组),理肺汤高、中、低剂量组(C、D、E组)、地塞米松组(F组)。复制哮喘模型,用药后肺组织病理HE染色,采用ELISA法测定血清中白介素(IL)-1β、IL-2、IL-13、IL-4、IL-5、肿瘤坏死因子-α(TNF-α)的含量;计算Th1、Th2类细胞总量及Th1/Th2比值。结果:镜下,A组未见炎性病理改变;B组可见典型支气管哮喘病理表现;治疗后C、F组对肺组织的炎性病理缓解最为明显;Th1类细胞因子变化,A组IL-1β、TNF-α血清含量低于其他各组(P<0.01,P<0.05);用药后C、D、E、F组IL-1β、TNF-α血清含量低于B组(P<0.01);A组IL-2血清含量高于B、D、E组(P<0.01);用药后C、F组血清IL-2含量较B组显著增加(P<0.01);Th2类细胞因子变化,A组IL-4、IL-5、IL-13血清含量明显低于其他各组(P<0.01);各用药组Th2类细胞因子血清含量明显低于B组(P<0.01);血清Th1/Th2比值可见,A组血清Th1/Th2比值高于B、D、E组(P<0.01);C、D、E、F组比值较B组升高(P<0.01)。结论:理肺汤高剂量组可以缓解肺组织病理损伤,并通过影响Th1、Th2类细胞因子血清含量,调节Th1/Th2比值失衡。
Objective: To observe the effects of LiFeiTang on lung pathological changes and serum Th1 / Th2 subsets imbalance in asthmatic rats. Methods: Forty eight male Sprague-Dawley rats were randomly divided into normal group (group A), model group (group B), high and low dose of Lifeng Tang (group C, D and E), dexamethasone group Group F). The model of asthma was duplicated and the histopathological changes were observed by HE staining. The levels of IL-1β, IL-2, IL-13, IL-4, IL-5 and TNF- ) Content; calculated Th1, Th2 type cells and Th1 / Th2 ratio. Results: In group A, no inflammatory pathological changes were found in group A; the pathological appearances of typical bronchial asthma were observed in group B; the inflammatory pathological changes of lung were the most obvious in groups C and F after treatment; the changes of Th1 cytokines, The serum levels of IL-1β and TNF-α in group C, D, E and F were lower than those in group B (P <0.01, P <0.01) (P <0.01); The content of IL-2 in group A and C was significantly higher than that in group B (P <0.01); The change of Th2 type cytokines, Serum levels of IL-4, IL-5 and IL-13 in group A were significantly lower than those in other groups (P <0.01); serum levels of Th2 cytokines in each group were significantly lower than those in group B (P <0.01) The ratio of Th2 in group A was higher than that in group B, D and E (P <0.01). The ratio of group C, D, E and F was higher than that in group B (P <0.01). Conclusion: The high-dose LiFeiTang decoction can alleviate the pathological damage of lung tissue and regulate the imbalance of Th1 / Th2 ratio by affecting the serum levels of Th1 and Th2 cytokines.
