目的探讨转录调节因子HIF-1在非小细胞肺癌(NSCLC)中的表达水平及其与PTEN关系。方法采用(SP)免疫组织化学方法检测60例NSCLC及40例癌旁正常肺组织中HIF蛋白、PTEN基因的表达,并分析两者相关性。结果 NSCLC组织中HIF-1阳性率75.0%,明显高于癌旁正常肺组织,PTEN在非小细胞肺癌组织中的阳性率为41.54%,显著低于非肺癌组织的阳性率88.89%(P<0.05);HIF和PTEN两者表达强度之间呈显著负相关(r=0.560 2,P<0.01)。结论 NSCLC组织存在HIF-1蛋白的过表达,PTEN蛋白的表达缺失在肺癌发生中可能起重要作用,并可能影响肿瘤的分化和预后,联合测定HIF-1和PTEN作为临床早期判断NSCLC侵袭、转移的指标之一。
Objective To investigate the expression of transcription factor HIF-1 in non-small cell lung cancer (NSCLC) and its relationship with PTEN. Methods The expression of HIF protein and PTEN gene in 60 cases of NSCLC and 40 cases of adjacent normal lung tissues were detected by (SP) immunohistochemistry. The correlation between them was analyzed. Results The positive rate of HIF-1 in NSCLC tissues was 75.0%, which was significantly higher than that in adjacent normal lung tissues. The positive rate of PTEN in non-small cell lung cancer tissues was 41.54%, which was significantly lower than that in non-lung cancer tissues (88.89%, P < 0.05). There was a significant negative correlation between the expressions of HIF and PTEN (r = 0.560 2, P <0.01). Conclusions The overexpression of HIF-1 protein in NSCLC tissues may play an important role in the development of lung cancer, which may affect the differentiation and prognosis of NSCLC. The combination of HIF-1 and PTEN can be used as an early diagnosis of NSCLC invasion and metastasis One of the indicators.
