目的评价Beagle犬自体子宫-卵巢移植动物模型构建的可行性。方法选择成年Beagle犬行子宫、输卵管、卵巢联合摘取,离体15~30min再行自体原位移植。移植后3周剖腹探查子宫、卵巢存活情况。并对子宫存活的Beagle犬采用药物诱导发情,观察发情行为及检测阴道脱落上皮细胞学的变化。结果8只犬,有6只犬血管吻合及自体移植手术成功,移植后血管短时全部畅通。手术当天犬死亡2只,分别死于腹腔内出血、失血过多。2只犬分别存活3、28d,另2只犬长期存活,移植子宫、卵巢成活良好。1只子宫长期存活犬药物诱导发情后,阴道上皮细胞受卵巢雌激素影响而发生变化,移植后7个月自然受孕并经阴道分娩。结论Beagle犬自体子宫卵巢移植模型的建立是可行的,存活子宫附件可自行妊娠与分娩,这将为人类子宫移植提供相关实验依据。
Objective To evaluate the feasibility of constructing an autologous uterine-ovarian transplantation animal model in Beagle dogs. Methods Adult Beagle dogs were selected for uterus, fallopian tube and ovary extraction, and then autologous orthotopic transplantation in vitro for 15 ~ 30 minutes. 3 weeks after transplantation laparotomy exploration of the uterus, ovarian survival. Uterine survival of Beagle dogs induced by drug-induced estrus, estrus behavior and detection of vaginal epithelial cells change. Results 8 dogs and 6 dogs were successful in vascular anastomosis and autotransplantation. Two dogs died on the day of surgery, respectively, died of intra-abdominal bleeding, excessive bleeding. Two dogs survived 3,28 days, respectively. The other two dogs survived long term, and the uterus was transplanted. The ovaries survived well. A long-term survival of the uterus after drug-induced estrus vaginal epithelial cells affected by ovarian estrogen changes occur after 7 months of natural pregnancy and vaginal delivery. Conclusion The establishment of the model of autologous uterine ovary transplantation in Beagle dogs is feasible. The surviving uterine attachment can be used for pregnancy and childbirth, which will provide relevant experimental evidence for human uterine transplantation.
