目的观察寿胎丸对去势大鼠血清雌激素水平和骨形态计量学参数的影响。方法 84只雌性SD大鼠随机分为6组,除假手术组外,其余5组建立去势模型(均行双侧卵巢切除术),寿胎丸高、中、低剂量组每天灌胃剂量分别为26.8,13.4,6.7 g·kg-1的药液,阳性对照药戊酸雌二醇组灌胃剂量为0.104 mg·kg-1的药液,假手术组与模型组予等量生理盐水灌胃。于术后12周处死动物,取静脉血检测血清雌二醇(E2)水平;获取右侧胫骨标本测定骨形态计量学参数。结果与假手术组比较,模型组血清E2水平下降且差异具有统计学意义(P<0.05);与模型组比较,寿胎丸高、中、低剂量组及戊酸雌二醇组血清E2水平均有明显升高(P<0.05);寿胎丸高、中剂量组及戊酸雌二醇组血清E2水平与寿胎丸低剂量组比较差异均有统计学意义(P<0.05)。与假手术组及模型组比较,各给药组在骨矿化沉积率无明显变化(P>0.05);与模型组比较,各给药组骨小梁面积百分数、骨小梁厚度、骨小梁密度、骨表面骨形成率、骨体积骨形成率升高,骨小梁分离度及破骨细胞数则降低,差异均有统计学意义(P<0.05)。血清E2水平与骨形态计量学中骨小梁面积百分数、骨小梁厚度、骨小梁密度骨小梁分离度、骨表面骨形成率、骨体积骨形成率、破骨细胞数明显相关(P<0.05)。结论雌激素水平与绝经后骨质疏松骨组织的微观结构改变密切相关。寿胎丸中、高剂量可提高血清E2水平,改善骨组织微观结构、抑制骨结构退化、促进骨形成、抑制骨吸收。
Objective To observe the effect of Shoutai Pill on serum estrogen level and bone metrology parameters in ovariectomized rats. Methods 84 female SD rats were randomly divided into 6 groups. Except the sham-operated group, castration was performed in the remaining 5 groups (both ovariectomies were performed). The daily dose of ShouTaiWan high, middle and low dose group Respectively, 26.8,13.4,6.7 g · kg-1 of the liquid, the positive control group of estradiol valerate injection intragastric dose of 0.104 mg · kg-1 of the liquid, sham-operated group and model group were given the same amount of normal saline Gavage. The animals were sacrificed at 12 weeks after operation, and the serum estradiol (E2) level was measured by venous blood. The right tibia specimens were obtained for bone morphometry. Results Compared with the sham-operation group, the level of serum E2 decreased and the difference was statistically significant (P <0.05). Compared with the model group, serum E2 level of high, medium and low dose ShouTaiWan group and estradiol valerate group (P <0.05). The serum E2 level of ShouTaiWan high and middle dose group and estradiol valerate group was significantly lower than that of Shoutai Wan low dose group (P <0.05). Compared with sham operation group and model group, there was no significant change in the rate of bone mineralization in each group (P> 0.05). Compared with the model group, the trabecular area percentage, trabecular thickness, Beam density, bone surface bone formation rate, bone volume bone formation rate increased, trabecular separation and osteoclast number decreased, the differences were statistically significant (P <0.05). Serum E2 levels were significantly associated with the percentage of trabecular area, trabecular thickness, trabecular bone trabecular separation, bone surface formation rate, volumetric bone formation rate and osteoclast number in bone histomorphometry (P <0.05). Conclusion The estrogen level is closely related to the changes of microstructure in postmenopausal osteoporotic bone tissue. Shou fetal pill, high dose can increase serum E2 levels, improve the microstructure of bone tissue, inhibit bone structure degradation, promote bone formation, inhibit bone resorption.
