[目的]提高枯草芽孢杆菌抗菌物质浓缩回收效率。[方法]通过单因素和正交试验,对发酵代谢产物进行超滤浓缩工艺优化。[结果]使用截留分子质量为3 k D的超滤柱、超滤温度40℃、p H值8.5、压力0.10 MPa为最佳超滤浓缩工艺。该条件下,膜通透量可以达到239.69 m L/(min·cm2),蛋白截留率90.2%,浓缩倍数为1.27倍,浓缩液抑菌活性是原来的1.3倍。[结论]通过超滤浓缩工艺优化,显著提高了BSD-2抗菌物质的回收效率,为产业化生产奠定基础。
[Objective] The research aimed to improve the concentration and recovery efficiency of Bacillus subtilis antimicrobial substance. [Method] Through the single factor and orthogonal experiment, the fermented metabolites were optimized by ultrafiltration concentration. [Result] The optimal ultrafiltration concentration was obtained by using ultrafiltration membrane with molecular weight cut off of 3 kD, ultrafiltration temperature of 40 ℃, pH value of 8.5 and pressure of 0.10 MPa. Under these conditions, the membrane permeability reached 239.69 m L / (min · cm2), the protein rejection rate was 90.2% and the concentration ratio was 1.27 times. The antibacterial activity of the concentrate was 1.3 times that of the original one. [Conclusion] The optimization of ultrafiltration concentration technology significantly improved the recovery efficiency of BSD-2 antibacterial substance and laid the foundation for industrial production.
