以北柴胡(Bupleurum chinense DC.)种内杂交所得的F1代96株植株为作图群体,利用拟测交理论,进行北柴胡遗传图谱构建。经父母本多态性筛选,从30条ISSR(inter-simple sequence repeat,内部简单重复序列)和44对SSR(simple sequence repeat,简单重复序列)引物中筛选出28条ISSR和14对SSR多态性引物。对F1代进行基因型和连锁分析,初步构建了包含13个连锁群、80个(72个ISSR和8个SSR)位点的首张北柴胡遗传图谱。该图谱覆盖长度2633.9cM,平均图距33.4cM,13个连锁群包含2~31个标记不等,连锁群遗传距离15.4~1295.7cM。该图谱为北柴胡性状基因定位、图位克隆以及分子标记辅助选择育种等研究奠定了基础。
Ninety-six F1 plants derived from a crossbreed of Bupleurum chinense DC. Were used as a mapping population. The genetic map of Bupleurum chinense DC was constructed based on the theory of quasispecies. A total of 28 ISSRs and 14 pairs of SSR polymorphisms were screened from 30 ISSR primers and 44 pairs of simple sequence repeats (SSR) Sex primers. Genotype and linkage analysis of F1 generation was carried out, and the first map of North Bupleurum containing 13 linkage groups, 80 (72 ISSRs and 8 SSRs) loci was constructed. The map covered a length of 2633.9cM with an average distance of 33.4cM. There were 2 ~ 31 markers in 13 linkage groups and a genetic distance of 15.4 ~ 1295.7cM in linkage group. The map laid the foundation for the research on the characters of Beachariar Bupleurum trait loci, map cloning and molecular marker assisted selection breeding.
