目的探讨孕期及哺乳期低剂量三丁基锡(TBT)暴露对子代昆明(KM)小鼠生长发育的影响。方法将清洁级健康成年妊娠昆明小鼠随机分为100、10、1μg/kgTBT染毒组和阴性对照组(玉米油),每组6只。从妊娠第6天(GD6)起,采用经口灌胃方式进行染毒,每天1次,直至哺乳期结束,灌胃剂量按照每天称量的体重计算。仔鼠出生后记录仔鼠数量,雌、雄数量及体重,仔鼠出生后第1、3、5、7、14、21天时称量仔鼠体重,出生后第12天(PND12)开始观察仔鼠睁眼情况,出生后第13天开始观察睾丸下降情况,出生后第20天开始观察阴道张开情况。结果100μg/kg剂量组母鼠孕期体重净增加量显著降低(P<0.01),各染毒组母鼠哺乳期体重增加量、产仔数量及仔鼠雌、雄比例,差异无统计学意义(P>0.05)。10μg/kg剂量组仔鼠体重在出生后第1天和各染毒组仔鼠在出生后第3、5、21天比对照组显著降低(P<0.05,P<0.01),100μg/kg剂量组仔鼠出生后第1天到第3天体重增加量及10μg/kg和100μg/kg剂量组仔鼠在出生后第14到20天体重增加量比对照组显著降低(P<0.05,P<0.01)。各TBT染毒组仔鼠的阴道张开时间比对照组显著提前,睁眼时间、睾丸下降时间显著延迟(P<0.05,P<0.01)且随着染毒剂量的增加睁眼时间有延迟的趋势。结论孕期及哺乳期低剂量TBT暴露可以影响仔鼠的生长发育。
Objective To investigate the effects of low dose tributyltin (TBT) exposure during gestation and lactation on the growth and development of offspring Kunming (KM) mice. The vaginal opening time of the offspring of TBT exposure group was significantly earlier than that of the control group, the time of opening eyes and testicular descent was significantly delayed (P <0.05, P <0.01), and the opening time was delayed with the increase of exposure dose trend. Conclusion Exposure to low doses of TBT during pregnancy and lactation can affect the growth and development of pups.
