目的探讨Hes1和Jagged1在胃癌中的表达及临床意义。方法应用免疫组织化学SP-9000法及实时荧光定量(qRT-PCR)法检测26例胃癌组织和20例癌旁正常组织中Hes1和Jagged1的表达水平及相应Hes1mRNA和Jagged1mRNA的表达情况,分析其与胃癌各临床因素之间的关系。结果 Hes1在胃癌组织中的阳性表达率(57.7%)高于癌旁正常组织(20.0%),差异有显著性(χ~2=5.161,P=0.023),而Jagged1在胃癌组织中的阳性表达率(34.6%)低于癌旁正常组织(80.0%),差异有显著性(χ~2=7.645,P=0.006)。胃癌中Hes1mRNA的表达量(1.12±0.11)明显高于癌旁正常组织(0.11±0.02),而癌旁正常组织中Jagged1mRNA的表达量(4.76±1.92)明显高于胃癌组织(1.02±0.09)。Hes1的表达与胃癌的浸润深度、分化程度、脉管浸润密切相关(P<0.05),与年龄、性别、临床分期及有无淋巴结转移无关(P>0.05)。Jagged1的表达与胃癌的浸润深度、分化程度、脉管浸润密切相关(P<0.05),与年龄、性别、临床分期及有无淋巴结转移无关(P>0.05)。胃癌组织中Hes1和Jagged1两者的表达呈负相关(r=-0.862,P=0.000)。结论 Hesl和Jagged1的异常表达与胃癌的发生、浸润及转移密切相关,检测两种蛋白及mRNA定量对于胃癌的早期诊断及预后具有一定的参考价值。
Objective To investigate the expression and clinical significance of Hes1 and Jagged1 in gastric cancer. Methods The expression of Hes1 and Jagged1 and the expression of Hes1 mRNA and Jagged1 mRNA in 26 cases of gastric cancer tissues and 20 cases of normal tissues adjacent to cancer tissues were detected by SP-9000 and qRT-PCR. The expression of Jagged1 was closely related to the depth of invasion, differentiation and vascular infiltration of gastric cancer (P <0.05), but not to age, gender, clinical stage and lymph node metastasis (P> 0.05). The expression of Hes1 and Jagged1 in gastric cancer tissue was negatively correlated (r = -0.862, P = 0.000). Conclusions The abnormal expression of Hesl and Jagged1 is closely related to the occurrence, invasion and metastasis of gastric cancer. It is of certain value to detect the expression of both proteins and mRNA in the early diagnosis and prognosis of gastric cancer.
