目的观察小鼠皮肤着色真菌病模型发病过程中Th相关细胞因子基因表达的动态变化,并进一步探讨其发病机制。方法采用卡氏枝孢霉在ICR小鼠皮下接种建立皮肤着色真菌病模型,在不同时间点RT-PCR法检测皮损组织IL-10、IFN-γ的mRNA表达水平。结果皮损局部IL-10基因表达水平逐渐下降,免疫正常组低于免疫抑制组;相反IFN-γ基因表达水平逐渐上升,免疫正常组高于免疫抑制组。结论 Th1与Th2细胞克隆转向可能在着色真菌病发病中起重要作用。动态跟踪Th1与Th2反应的情况可能为皮肤着色真菌病的发展、转归提供有价值的参考。
Objective To observe the dynamic changes of Th-related cytokine gene expression during the development of skin pigmented mycosis model in mice and to explore its pathogenesis. Methods The model of skin pigmented mycosis was established by subcutaneously inoculation of C. Cardiomycetes in ICR mice. The mRNA expression levels of IL-10 and IFN-γ in skin lesions were detected by RT-PCR at different time points. Results The expression of IL-10 gene in the lesional lesion gradually decreased. The immunocompetent group was lower than the immunosuppressed group. On the contrary, the expression of IFN-γ gene was gradually increased, while the immunocompromised group was higher than the immunosuppressed group. Conclusion The cloning and turning of Th1 and Th2 cells may play an important role in the pathogenesis of pigmented fungal diseases. The dynamic tracking of the reaction between Th1 and Th2 may provide a valuable reference for the development and prognosis of skin pigmented mycosis.
