目的:研究十精丸含药血清对光老化人皮肤成纤维细胞(ESF-1)分泌MMP-1、TIMP-1的影响。方法:制备十精丸含药血清,采用UVA照射ESF-1细胞建立光老化模型,照射后用含不同浓度十精丸的大鼠血清进行干预,24h后采用ELISA方法检测各组细胞分泌MMP-1、TIMP-1的含量。结果:与正常对照组比较,模型对照组细胞活力降低、MMP-1表达升高、TIMP-1表达降低(P<0.01);与模型对照组比较,十精丸中、高剂量含药血清组细胞活力升高、MMP-1表达降低、TIMP-1表达升高(P<0.05,P<0.01)。结论:十精丸含药血清通过调节MMP-1、TIMP-1的表达来拮抗UVA对皮肤成纤维细胞的损伤。
Objective: To study the effect of decoction-containing serum on the secretion of MMP-1 and TIMP-1 by photoaging human dermal fibroblasts (ESF-1). Methods: The decoction-containing serum was prepared. The model of photo-aging was established by UVA irradiation of ESF-1 cells. After irradiation, the rats were treated with serum containing different concentrations of decoction. Serum MMP- 1, the content of TIMP-1. Results: Compared with the normal control group, the viability of the model control group decreased, the expression of MMP-1 increased and the expression of TIMP-1 decreased (P <0.01). Compared with the model control group, The cell viability increased, the expression of MMP-1 decreased and the expression of TIMP-1 increased (P <0.05, P <0.01). Conclusion: Decoction-containing serum can antagonize the injury of skin fibroblasts by regulating the expressions of MMP-1 and TIMP-1.
