目的:探讨吗啡(morphine,Mor)点燃条件位置性偏爱(conditioned place preference,CPP)重现大鼠海马CAl区N-甲基-D-天冬氨酸受体(NMDA受体)调节亚基NR2B,钙依赖钙调蛋白激Ⅱ(Ca~(2+)/calmodulin dependent protein kinase Ⅱ,CaMKⅡ)表达的变化。方法:用恒量法(10 mg·kg~(-1))给大鼠连续颈背部皮下注射(subcutaneous,sc)吗啡8 d建立CPP模型;用生理盐水替代吗啡训练大鼠10 d,使形成的CPP逐渐消退;单次sc2.5 mg·kg~(-1)吗啡点燃已消退的CPP。用免疫组化法检测吗啡点燃CPP重现大鼠海马CA1区NR2B、CaMKⅡ表达,图像分析系统测定阳性反应产物的平均灰度值的变化。结果:sc10 mg·kg~(-1)吗啡8 d建立CPP,生理盐水训练10 d使已形成的CPP消退,小剂量吗啡(2.5 mg·kg~(-1))使消退的CPP重现;吗啡点燃(CPP重现大鼠海马CA1区NA2B、CaMKⅡ表达,与对照组比较呈显著增加(P<0.05)。结论:小剂量吗啡诱发大鼠CPP重现行为可能与海马CA1区NR2B、CAMKⅡ表达增加有关。
OBJECTIVE: To investigate the effect of morphine (Mor) on conditioned place preference (CPP) in reproducing hippocampal CA1 N-methyl-D-aspartate receptor (NMDA receptor) regulatory subunit NR2B , Calcium dependent Ca ~ (2 +) / calmodulin dependent protein kinase Ⅱ (CaMKⅡ) expression. Methods: CPP model was established by subcutaneous injection of morphine (subcutaneous, sc) on the dorsal neck of the rats with a constant dose (10 mg · kg -1) for 8 d. Rats were trained with saline instead of morphine for 10 days. CPP gradually subsided; a single sc with 2.5 mg · kg -1 morphine ignited CPP. The expression of NR2B and CaMKⅡ in hippocampus CA1 of rats was detected by immunohistochemistry after morphine ignited CPP, and the average gray value of positive reaction products was measured by image analysis system. Results: CPP was established 8 days after sc 10 mg · kg -1 morphine treatment. CPP was regressed after 10 days of saline training and 2.5 mg · kg -1 morphine was regained. Morphine ignition (CPP) showed that expression of NA2B and CaMKII in CA1 area of hippocampus was significantly increased compared with control group (P <0.05) .Conclusion: CPP reappearance in low dose morphine rats may be associated with the expression of NR2B and CAMKII in hippocampal CA1 area Increase related.
