目的探讨TS、TP作为5-FU化疗敏感性的预测指标的可行性。方法选取我院2010年—2012年手术切除并经病理证实的大肠癌存档石蜡标本50例,术前均未进行过任何辅助治疗,术后均进行5-FU为主的化疗,检测50例大肠癌组织及20例正常大肠黏膜组织中TP、TS表达。联用100μ/ml的IFN-γ24h后2株细胞对5-FU敏感性增高。结果正常大肠黏膜组织TS水平高于大肠癌组织,而大肠癌组织TP水平高于正常组织。TS表达水平与大肠癌患者5-FU化疗预后呈负相关,与患者年龄、性别无关,与肿瘤大小、分化程度、浸润深度、淋巴结转移、Dukes分期有关。5-FU治疗后,TS表达增加,而联用IFN-γ后,TS表达降低。结论 IFN-γ可诱导大肠癌细胞TP表达增强、抑制TS表达,增强5-FU、5-DFUR抗肿瘤活性。
Objective To investigate the feasibility of using TS and TP as predictors of 5-FU chemosensitivity. Methods Fifty cases of paraffin-embedded specimens of colorectal cancer surgically and pathologically confirmed in our hospital from 2010 to 2012 were selected. No adjuvant therapy was given before operation. All patients underwent 5-FU-based chemotherapy after operation. 50 cases of large intestine The expression of TP and TS in cancer tissue and 20 normal colorectal mucosa tissues. When combined with 100μg / ml IFN-γ24h, the two cells were more sensitive to 5-FU. Results The TS of normal colorectal mucosa was higher than that of colorectal cancer, while the level of TP in colorectal cancer was higher than that of normal tissues. The expression of TS was negatively correlated with the prognosis of 5-FU chemotherapy in patients with colorectal cancer, which was not related to the age and gender of the patients. It was related to the tumor size, differentiation, depth of invasion, lymph node metastasis and Dukes staging. After treatment with 5-FU, the expression of TS was increased, while the expression of TS was decreased after IFN-γ treatment. Conclusion IFN-γ can induce the expression of TP in colorectal cancer cells, inhibit the expression of TS, and enhance the antitumor activity of 5-FU and 5-DFUR.
