目的:探讨不同修复条件及修复液对细胞角蛋白(广谱)单克隆抗体(Pan)的阳性表达程度的影响,筛选优势抗原修复方法。方法:对11例鼻腔鼻窦内翻性乳头状瘤(SNIP)组织石蜡切片,分别采用高温高压EDTA修复、微波EDTA修复、微波柠檬酸钠修复、高温高压柠檬酸钠修复,利用Image-Pro Plus图像分析仪对每张切片随机取5个视野,平均计算200个单位染色区域面积,测定染色区阳性反应的平均吸收度值。结果:高温高压柠檬酸钠修复、微波EDTA修复、微波柠檬酸钠修复与高温高压EDTA修复Pan的阳性表达分别为0.324±0.051、0.325±0.056、0.303±0.061及0.365±0.023;前3种方法与高温高压EDTA修复比较,均差异有统计学意义(均P<0.05)。结论:应用高温高压EDTA修复液可最大化增加Pan的阳性表达强度,达到最佳染色结果,是一种值得推广的免疫组织化学修复方法,可为临床工作提供更好的指导作用。
Objective: To investigate the effect of different repair conditions and repair solution on the positive expression of cytokeratin (Pan) monoclonal antibody (Pan), and to screen the method of antigen retrieval. Methods: Eleven cases of nasal sinus inverted papilloma (SNIP) tissue paraffin sections were treated with high-temperature high-pressure EDTA, microwave EDTA, microwave sodium citrate, and sodium pyrophosphate respectively. Image-Pro Plus The analyzer randomly selected 5 fields for each slice and calculated the area of 200 units on average and determined the average absorbance value of the positive reaction in the stained area. Results: The positive expressions of Pan in high temperature and high pressure sodium citrate, microwave EDTA, microwave sodium citrate and high temperature and high pressure EDTA were 0.324 ± 0.051,0.325 ± 0.056,0.303 ± 0.061 and 0.365 ± 0.023, respectively. The first three methods High-temperature high-pressure EDTA repair comparison, the difference was statistically significant (P <0.05). Conclusion: The application of high-temperature and high-pressure EDTA repair fluid can maximize the positive expression intensity of Pan and achieve the best staining results. It is a worthwhile promotion of immunohistochemical repair method, which can provide better guidance for clinical work.
