目的:通过研究茵栀黄注射液对TNF-α作用下大鼠原代肝细胞Mrp2表达的影响,分析茵栀黄注射液的利胆退黄作用机制。方法:三明治法培养大鼠原代肝细胞。以茵栀黄注射液和/或TNF-α处理大鼠原代肝细胞。以定量PCR和Western-blot检测大鼠原代肝细胞Mrp2 mRNA及蛋白的表达。结果:对照组肝细胞Mrp2 mRNA表达高于TNF组(0.568±0.056,P<0.05),低于茵栀黄组(1.591±0.107,P<0.05);茵栀黄加TNF组肝细胞Mrp2 mRNA(1.079±0.087)表达高于TNF组(P<0.05)。对照组肝细胞Mrp2蛋白(0.768±0.070)表达高于TNF组(0.119±0.031,P<0.05),低于茵栀黄组(1.895±0.129,P<0.05);茵栀黄+TNF组肝细胞Mrp2蛋白表达(1.118±0.091)高于TNF组(P<0.05)。结论:茵栀黄注射液能改善TNF-α所导致的大鼠原代肝细胞Mrp2表达下降。
OBJECTIVE: To study the effect of Yinzhihuang injection on the expression of Mrp2 in rat primary hepatocytes under the action of TNF-α, and to analyze the mechanism of Yinhuanghuang injection on the gallbladder jaundice. METHODS: Rat primary hepatocytes were cultured by sandwich method. Rat primary hepatocytes were treated with Yinzhihuang injection and/or TNF-α. The expression of Mrp2 mRNA and protein in rat primary hepatocytes was detected by quantitative PCR and Western-blot. Results: The expression of Mrp2 mRNA in hepatocytes of control group was higher than that of TNF group (0.568±0.056, P<0.05), which was lower than that of Yinzhihuang group (1.591±0.107, P<0.05). 1.079±0.087) expression was higher than TNF group (P<0.05). The expression of Mrp2 protein (0.768±0.070) in the liver of the control group was higher than that of the TNF group (0.119±0.031, P<0.05), lower than that of the Yinzhihuang group (1.895±0.129, P<0.05); the liver cells of the Yinzhihuang+TNF group Mrp2 protein expression (1.118±0.091) was higher than that of TNF group (P<0.05). Conclusion: Yinzhihuang injection can improve the expression of Mrp2 in rat primary hepatocytes induced by TNF-α.
