目的:观察喜树碱衍生物SN-38对人白血病细胞系HL-60细胞的增殖抑制和诱导凋亡作用,并探讨其可能的作用机制。方法:采用CCK法检测SN-38对HL-60细胞的增殖抑制作用;采用流式细胞术检测细胞凋亡率;特异性荧光底物检测Caspase-3、Caspase-8、Caspase-9活性;观察不同种类Caspase抑制剂对SN-38诱导HL-60细胞的增殖和凋亡作用的影响。结果:CCK法显示SN-38明显抑制HL-60细胞增殖,流式细胞凋亡分析及Caspase抑制剂研究,显示SN-38通过内源性Caspase信号通路诱导HL-60细胞凋亡。结论:SN-38通过内源性Caspase信号通路抑制HL-60细胞增殖和诱导凋亡。
OBJECTIVE: To observe the proliferation and apoptosis induced by the camptothecin derivative SN-38 on human leukemia cell line HL-60 and to explore its possible mechanism. Methods: The inhibitory effect of SN-38 on the proliferation of HL-60 cells was detected by CCK method. The apoptosis rate of HL-60 cells was detected by flow cytometry. The activity of Caspase-3, Caspase-8 and Caspase- Effect of Different Caspase Inhibitors on the Proliferation and Apoptosis of HL-60 Cells Induced by SN-38. Results: CCK assay showed that SN-38 significantly inhibited the proliferation of HL-60 cells. Flow cytometry analysis and Caspase inhibitor study showed that SN-38 induced HL-60 cell apoptosis through endogenous Caspase signaling pathway. Conclusion: SN-38 inhibits the proliferation and induces apoptosis of HL-60 cells through endogenous Caspase signaling pathway.
