[目的]观察中药正肝方药物血清对甲胎蛋白(AFP)刺激的AFPsiRNA-HepG2细胞增殖的影响。[方法]正肝方浸膏粉经灌胃给药制备正肝方大鼠血清,体外培养AFPsiRNA-HepG2细胞,将细胞分为5%正常大鼠血清、5%正肝方血清、10%正常大鼠血清、10%正肝方血清、5%正常大鼠血清加AFP刺激、5%正肝方血清加AFP刺激、10%正常大鼠血清加AFP刺激、10%正肝方血清加AFP刺激共8组,MTT法检测细胞增殖。[结果]无AFP刺激时,正肝方血清组与正常大鼠血清组OD值差异无统计学意义(P>0.05);有AFP刺激时,正肝方血清组与正常大鼠血清组OD值差异有统计学意义(P<0.01),正肝方血清组显著抑制AFPsiRNA-HepG2细胞增殖。[结论]正肝方血清通过影响AFP介导的AFPsiRNA-HepG2细胞的增殖抑制肝癌细胞增殖。
[Objective] To observe the effect of Zhengganfang serum on the proliferation of AFP-stimulated AFPsiRNA-HepG2 cells. [Method] AFPsiRNA-HepG2 cells were prepared by intragastric administration of Zhengganfang extract powder by intragastric administration. AFPsiRNA-HepG2 cells were cultured in vitro. The cells were divided into 5% normal rat serum, 5% Zhengganfang serum and 10% normal Serum of rat, 10% of Zhenggan serum, 5% of normal serum plus AFP stimulation, 5% of Zhenggan Liver serum plus AFP stimulation, 10% of normal rat serum plus AFP stimulation, 10% of Zhenggan Liver serum plus AFP stimulation A total of 8 groups, MTT assay cell proliferation. [Results] There was no significant difference in the OD value of serum of Zhengganfang serum group and normal rat without AFP stimulation (P> 0.05). When AFP was stimulated, the serum OD value of Zhengganfang serum group and normal rat The difference was statistically significant (P <0.01), Zhenggan Fang serum group significantly inhibited the proliferation of AFPsiRNA-HepG2 cells. [Conclusion] Zhengganfang serum can inhibit the proliferation of hepatoma cells by affecting the proliferation of AFP-mediated AFPsiRNA-HepG2 cells.
