目的:研究虫草肾茶胶囊对培养在高糖条件下系膜细胞(HMC)增殖及细胞上清液中胶原纤维(Col-IV)的表达影响,探讨其在糖尿病肾病(DN)防治中的意义。方法:利用四甲基偶氮唑蓝(MTT)比色法动态观察各种处理因素的作用下各组HMC增殖情况;酶联免疫吸附试验(ELISA)测定经体外培养的人肾小球系膜细胞在各种处理因素的作用下上清液中IV型胶原的表达情况,并以福辛普利为对照。结果:高糖能明显刺激体外培养的HMC增殖,同时促进HMC分泌Col-IV(P<0.05或P<0.01)。而虫草肾茶胶囊能抑制HMC增殖及HMC分泌Col-IV,并呈一定量效关系(P<0.05或P<0.01),且某些方面明显优于福辛普利。结论:高糖可促进HMC增殖及Col-IV分泌,虫草肾茶胶囊能明显抑制高糖条件下HMC增殖及其分泌Col-IV,从而发挥预防肾小球纤维化及硬化的发生、发展,进而延缓DN的进展。
Objective: To study the effect of Cordyceps and Renshen capsule on the proliferation of mesangial cells (HMC) cultured in high glucose conditions and the expression of collagen fibers (Col-IV) in cell supernatants, and to explore its significance in the prevention and treatment of diabetic nephropathy (DN). . METHODS: MTT colorimetry was used to observe the proliferation of HMC under the action of various treatment factors. The enzyme-linked immunosorbent assay (ELISA) was used to determine the human mesangial cells cultured in vitro. The expression of type IV collagen in the supernatant of cells under the action of various treatment factors was compared with fosinopril. RESULTS: High glucose significantly stimulated the proliferation of HMC cultured in vitro and promoted the secretion of Col-IV by HMC (P<0.05 or P<0.01). In addition, Cordyceps and Kidney Tea Capsules inhibited HMC proliferation and HMC secretion of Col-IV, and showed a certain dose-effect relationship (P<0.05 or P<0.01), and in some aspects were significantly better than fosinopril. Conclusion: High glucose can promote the proliferation of HMC and the secretion of Col-IV. Chongcao Shencha Capsule can inhibit the proliferation of HMC and the secretion of Col-IV under high glucose conditions, so as to prevent the occurrence and development of glomerular fibrosis and sclerosis. Delay the progress of DN.
