目的 :研究鼻咽癌细胞的调节性容积回缩 (RVD)及Cl-在其中所起的作用。方法 :活细胞图像分析低分化鼻咽癌细胞 (CNE - 2Z)在细胞外低渗刺激时的细胞容积改变 ,用离子置换法和通道阻断法阐明Cl-在RVD中所起的作用。结果 :细胞外低渗刺激使细胞膨胀并诱发RVD。在 16 0 - 2 30mOsmol/L范围内 ,RVD与细胞膨胀程度正相关。用葡萄糖酸取代灌流液Cl-时RVD增强 ,细胞内Cl-耗竭时RVD消失。氯通道阻断剂阻抑RVD ,但不同的阻断剂对RVD的阻抑率有明显差异。结论 :Cl-是CNE - 2Z细胞RVD的关键离子 ,Cl-通道激活 ,Cl-外流是RVD的主要机制。
AIM: To investigate the role of regulatory volume retraction (RVD) and Cl- in nasopharyngeal carcinoma cells. Methods: The changes of cell volume in poorly differentiated nasopharyngeal carcinoma cells (CNE - 2Z) under extracellular hypotonic stimulation were analyzed by living cell images. The role of Cl - in RVD was elucidated by ion exchange and channel blockade. Results: Extracellular hypotonic stimulation inflates the cells and induces RVD. In the range of 16 0-2 30 mOsmol / L, RVD was positively correlated with the degree of cell swelling. RVD was enhanced when gluconic acid was used instead of Cl- in perfusate and RVD disappeared when intracellular Cl-depletion occurred. Chloride channel blockers inhibit RVD, but different blockers have significant differences in RVD inhibition rate. Conclusion: Cl - is the key ion of RVD in CNE - 2Z cells. Cl - channel activation and Cl - efflux are the main mechanisms of RVD.
