目的:通过代谢组学研究方法,寻找大鼠寒凝证潜在的生物标志物;研究热性中药川芎对其潜在生物标志物的影响,探讨川芎治疗寒凝证的作用机制。方法:根据寒凝证的临床表现,制造寒凝血瘀大鼠模型;利用UPLC/Q-TOFMS BPI离子流图谱,分别绘制空白组、血瘀模型组、川芎大小剂量给药组的血浆BPI离子流指纹图谱,对图谱中分子离子峰进行分析,通过Markerlynx软件进行色谱峰自动识别和峰匹配,采用主成分分析法(PCA)对不同组间的数据进行模式识别,从而找到寒凝血瘀证潜在生物标志物及药物的作用靶点,阐明其作用机制。结果:找到寒凝血瘀证潜在生物标志物有N-甲基花生四烯酰胺、二氢神经鞘氨醇、N-丙基花生四烯酰胺、花生四烯酰基溶血卵磷脂、亚油酰基甘油磷脂酰胆碱、十六烷酸溶血磷脂酰胆碱、油酰甘油磷酯酰胆碱、十六烷基甘油磷脂酰胆碱8种。结论:热性药川芎对寒凝血瘀证大鼠的作用机制,可能是通过抑制使磷脂酰胆碱转化为花生四烯酸的磷脂酶A2,使血栓烷的合成水平降低,影响磷脂酰胆碱代谢途径,使血瘀症状减轻而发挥治疗作用的。
OBJECTIVE: To search potential biomarkers of cold sink coagulation in rats through metabonomics research, to study the effect of Chuanxiong on its potential biomarkers, and to explore the mechanism of action of Rhizoma Chuanxiong on treating cold syndrome. Methods: According to the clinical manifestations of cold syndromes, a cold-blooded blood stasis rat model was made. The UPLC / Q-TOFMS BPI ion chromatogram was used to draw the plasma BPI ion current of blank group, blood stasis model group and Chuanxiong size dose group Fingerprinting, analysis of molecular ion peaks in the spectrum, automatic identification and peak matching of chromatographic peaks by Markerlynx software, pattern recognition of data of different groups by principal component analysis (PCA) to find potential biomarkers of cold coagulation and blood stasis Markers and drug targets, clarify its mechanism of action. Results: The potential biomarkers of cold coagulation and blood stasis were N-methyl arachidonamide, dihydrosphingosine, N-propyl arachidonic acid amide, arachidonic acid lysolecithin, linoleyl glycerophospholipid Acetylcholine, palmitic acid lysophosphatidylcholine, oleoyl glycerol phosphatidylcholine, cetyl glycerophosphatidylcholine 8 kinds. CONCLUSION: The mechanism of action of Chuanxiong Huanxiong on rats with cold-coagulation and blood-stasis syndrome may be that the synthesis level of thromboxane may be reduced by inhibiting the phospholipase A2 that converts phosphatidylcholine into arachidonic acid, affecting the effect of phosphatidylcholine Metabolic pathways, to reduce the symptoms of blood stasis and play a therapeutic role.
